Objective: To study emodin on P2X2 / 3 receptor-mediated analgesic effect of neuropathic pain , and to explore its possible mechanism . Methods : Preparation of rat sciatic nerve chronic constriction injury (Chronic constriction injury, CCI) model of neuropathic pain , male rats were randomly divided into control group (Control), sham operation group (Sham), CCI rat model group (CCI ) , emodin treated control group (Emodin), CCI model emodin treatment group (CCI emodin) 5 groups. 1 , ethology methods rats were observed in five of the mechanical paw withdrawal reflex threshold (MWT) and thermal withdrawal reflex latency (TWL), observe the behavior of emodin on the impact of pain in CCI rats ; 2 , in situ hybridization and RT-PCR was observed in each group L4-L5 dorsal root ganglion P2X2 / 3 receptor mRNA expression 3 , immunohistochemical staining and Western Blotting observed the L4-L5 dorsal root ganglion P2X2 / 3 by protein expression . Results: 1 after surgery 5-7 d, rat model of neuropathic pain sciatic basic formation , CCI CCI emodin rat model group and the right hind limb of rats MWT and TWL significantly reduced, and the Control group , Sham group and Emodin group was statistically significant (p lt; 0.05), while the Sham group , Emodin comparable between groups, and Control was no significant difference (p gt; 0.05); surgery 7-14 d, CCI emodin rats MWT and TWL significantly increased, compared with CCI rats were significantly different (p lt; 0.05), compared with the Control groups had no significant difference (p gt; 0.05); 2, after 14 d, in situ hybridization , RT-PCR shows , P2X2 / 3 receptor mRNA in CCI rats L4-L5 dorsal root ganglia were significantly increased (p lt; 0.01), and in the CCI emodin rat dorsal root ganglia lower than the CCI group (p lt; 0.01); 3, immunohistochemistry and Western blot results showed that with the Control group , Sham group and Emodin group , CCI rats L4-L5 dorsal root ganglion P2X2 / 3 receptors protein expression was significantly increased (p lt; 0.01), while the CCI emodin group of dorsal root ganglion P2X2 / 3 receptor protein expression was lower than the CCI (p lt; 0.01). CONCLUSION : Emodin on primary sensory neurons P2X2 / 3 receptor-mediated neuropathic pain with inhibition.
