Inhibitory Effect of NDGA on Adenoid Cystic Carcinoma

Objective: To find potential chemotherapy for salivary adenoma cystic carcinoma, we study the expression of lipoxygenase in human salivary adenoma cystic carcinoma and the role of lipoxygenase in adenoma cystic carcinoma growth, the effect of lipoxygenase inhibitor NDGA. Methods: Cell proliferation was determined using CCK-8 colorimetric assay. Apoptosis was detected by Flow Cytometry and electron microscope. mRNA expression was assayed by RT-PCR. Cell lipoxygenase protein was detected by western blot, and the expression of subtype lipoxygenase was tested by EnVision immonohistochemistry. Gland carcinoma and lung metastatic animal model were established by injection of ACC-M cells to the nude BalB/C mice subcutaneouly and tail vein seperatly. Results: Lipoxygenase is expressed in human salivary adenoid cystic carcinoma with the high exprssion of 15-lipoxygenase. Lipoxygenase inhibitor NDGA can dose and time dependently inhibits adenoid cystic carcinoma growth through blocking cell cycle and modulating gene expression related to apoptosis. Conclusion: Lipoxygenase may play a role in the occurance of salivary adenoid cystic carcinoma, and lipoxygenase inhibitor may have potential chemotherapical value.
