Abstract


A series of indoor trials were conducted to investigate effects of different dissolved oxygen levels and exposure in air under different temperature on nonspecific immunity, metabolisitic enzymes activities of Apostichopus japonicus. The primary results were listed below.1 Effect of temperature and specifications on body wall respiration of Apostichopus japonicusThe body wall respiration of Sea cucumber (Apostichopus japonicus) was studied by using experimental ecological approach in this experiment. The results showed that the oxygen consumption rate significantly increased when the temperature rose up, and the oxygen consumption rate of the small-sized was much higher than that of large-sized specification.2 Effect of different dissolved oxygen levels and specifications on oxygen consumption rate of Apostichopus japonicusThe effect of different dissolved oxygen levels (1,3,5 and pure oxygen-saturated) and specifications on oxygen consumption rate of the muscle of Sea cucumber (Apostichopus japonicus) was studied by using experimental ecological approach in this experiment. The results showed that oxygen consumption rate of the small-sized wsa much higher than that of the large-sized specification, and the oxygen consumption rate increased with the increase of dissolved oxygen. But there is no obvious difference between the oxygen-saturated group and the control group.3 Effect of different dissolved oxygen levels on non-specific immune parameters of the body wall of Apostichopus japonicusThe effect of different dissolved oxygen levels (1,3,5 and 7mg / l) on non-specific immune parameters of the body wall of Sea cucumber (Apostichopus japonicus) was studied by using experimental ecological approach in this experiment. The results showed that there were different response between the body wall hydrolytic enzymes and antioxidant indicators under hypoxia stress: (1) the activity of the body wall acid phosphatase (ACP) and alkaline phosphatase (AKP) was significantly lower than the control group under hypoxia stress, showing that the enzyme activity of sea cucumber hydrolytic system was significantly inhibited by hypoxia stress, and significantly inhibited with the increas of stress; (2) the activity of superoxide dismutase and body wall enzyme (SOD), catalase (CAT) and total antioxidant (T-AOC) and the contents of malondialdehyde trace (MDA) and other parameters of antioxidation were first increased significantly with the increase of stress level under hypoxia stress, and declined afterwards. During the 72h in this study, the parameter of antioxidant on body wall eventually recovered to control level under the dissolved oxygen level of 5mg / l. However, under dissolved oxygen levers of 1 and 3mg / l, the related indicators were still significantly lower than the control level. This study showed that the activity of ACP, AKP, SOD, CAT and T-AOC and MDA content in the body wall are sensitive to the dissolved oxygen level of water. These parameters can be used to indicate the level of non-specific immunity of sea cucumber under hypoxia stress.4 Effect of different dissolved oxygen levels on metabolic enzymes activities of the body wall of Apostichopus japonicusThe effect of different dissolved oxygen levels (1,3,5 and 7mg / l) on metabolic enzymes activities of the muscle of Sea cucumber (Apostichopus japonicus) was studied by using experimental ecological approach in this experiment. The results showed that there were different response of the metabolic enzymes under hypoxia stress: (1) the hypoxia stress had a positive effect on the glycolysis enzymes, Hexokinase(HK)and Pyruvate kinase(PK) in a short time. (2) while it had a significantly negative effect on aerobic enzyme malate dehydrogenase(MDH) . (3) the hypoxia stress can fiercely increase the anaerobic enzyme avtivity. The lactate dehydrogenase(LDH) activity showed a dramaticaly rise during the experiment, and lactic acid(LD) contents showed a continuous accumulation.
