Vitiligo is a skin intractable due to epidermal melanocytes disappear or deletions caused by localized or generalized depigmentation of skin diseases, the global incidence rate of 0.1-2%. Although vitiligo is not a deadly disease, but it is easy to bring a heavy psychological burden on the patient and induce other skin diseases. The use of biocompatible membrane material melanocytes load and transfer for transplantation in the treatment of vitiligo, improve the success rate of treatment provides a new way. However, this method requires the use of membrane materials to meet the melanocyte cultures at the same time both to meet the required mechanical strength and transplanted to promote wound healing, so the selection and preparation of the appropriate membrane material challenging. The topics to have good biocompatibility of chitosan, gelatin and polyethylene glycol as a substrate by physical crosslinking of sodium sulfate were prepared chitosan, chitosan / gelatin and chitosan / poly glycol crosslinked composite membrane, and cross-linked film material's physical and mechanical properties were studied, and the composition and structure of the substrate on melanocyte morphology, proliferation and biological activity are discussed. The experimental results showed that: chitosan cross-linked film with good physical and mechanical properties and biocompatibility, support posts with melanocytes, and maintaining cell growth and proliferation activity, animal experiments show that the load on the film melanocytes able to xenograft metastases and expression activity, and can promote wound healing; chitosan / gelatin composite membranes crosslinked water absorption and water vapor transmission rate and improve the use of gelatin, cultured melanocytes have a better the shape and activity of the melanocytes - chitosan / gelatin matrix composite membrane complex for mice transplanted skin, melanocytes can successfully transferred into nude wound, forming a continuous layer melanocytes and having activity; Chitosan / PEG complex cross-linked film in the amount of PEG introduced to improve the hydrophilicity of the membrane and the water absorption and water vapor permeability, but also help to improve the morphology of melanocytes. The highlight of the subject membrane materials were prepared by physical cross-linking methods, to improve the mechanical properties of the material, meet the load cell, culture, transfer and transplant requirements. Meanwhile, the prepared membrane materials during migration both can play the role of protecting and supporting cells, but also in cell culture, the process of transfer and transplant melanocytes provide nutrients, the material itself but also play to accelerate wound healing effect is expected to be used for transplantation in the treatment of vitiligo.
