Abstract


Research background with systemic lupus erythematosus (SLE) is an autoimmune disease of multiple organ damage. By the onset of the main mechanism is the body's own immune tolerance destruction, a lot of organizing itself become the target antigen, leading to the production of autoantibodies and organ tissue damage. CD4 CD25 regulatory T cells (Treg) are key to the peripheral immune tolerance, excessive apoptosis of such cells is one of the pathogenic mechanisms of SLE [1]. Treg regulate important cells of the immune response as a group, which itself exists an adjustment process. Apoptosis is an important mechanism in the homeostatic regulation of immune cells in the immune response process and the fas / fasL system is the most important in the signaling pathways mediated apoptosis of death signaling molecules [2]. Foxp3 (forkhead wing helix transcription factor) in the CD4 CD25 Treg-specific expression, and play an important role in the development and function of the cell population, Foxp3 certain extent reflect CD4 CD25 Treg levels and activity [3]. various subtypes. SLE patients by flow cytometry in order to reduce the impact on the results of other subtypes, this study peripheral Foxp3 CD4 CD25 Treg expression of cell surface fas investigate the the fas apoptotic signal pathway in SLE patients of Foxp3 CD4 CD25 Treg abnormal apoptosis and the role of the mechanism of disease activity. Objective To investigate the the fas apoptosis signal transduction pathways in SLE patients Foxp3 CD4 CD25 Treg role in the abnormal apoptosis. The research method selected 25 cases of patients with active SLE remission 20 cases of SLE patients and healthy controls, 25 patients for the study, to detect all objects Foxp3 CD4 CD25 Treg surface fas expression in peripheral blood CD4 CD25 T cells Foxp3 expression. And fas expression of Foxp3 expression rate of disease activity (SLEDAI score). Results, peripheral Foxp3 CD4 CD25 Treg on the expression of the fas: active SLE group (23.72 ± 2.35)%, remission SLE group (14.0 ± 2.07)% in the healthy control group (10.13 ± 1.21)%, active SLE group than in the remission group (P = 0.003) and the control group (P lt; 0.001), to ease the group with the control group, the difference had no statistical significance, expression of fas expression on Foxp3 CD4 CD25 Treg SLEDAI score was positively correlated (r = 0.336, P = 0.024). 2 of Foxp3 expression in peripheral blood CD4 CD25 T cells: active SLE group (2.83 ± 0.30)%, remission of SLE group (5.38 ± 0.63)% healthy control group (8.12 ± 0.70)%. Activity in patients with peripheral blood CD4 CD25 T cells Foxp3 expression was significantly lower than patients in remission (P = 0.008) and the control group (P lt; 0.001); and remission also lower than that of the control group (P = 0.04). Peripheral blood Foxp3 expression and SLEDAI score was negatively correlated (r = -0.581, P lt; 0.001). Foxp3 CD4 CD25 Treg excessive apoptosis mediated by fas research conclusions SLE patients, which is one of the mechanisms that lead to SLE disease activity.
