Abstract


Objective : To investigate the in vitro isolation and amplification of CD4 CD25 T cells. Methods: immunomagnetic beads (MACS) sorting to obtain CD4 CD25 T cells ; using IL-2 (200IU / m) anti-CD3/CD28/beads right after the separation of CD4 CD25 T cells in vitro amplification . Results: 1, using MACS separation of adult peripheral blood CD4 CD25 T cells , purity of up to 81% -91 % . 2, IL-2 (200IU / m) anti-CD3/CD28/beads amplification method can achieve a certain degree of amplification results. Conclusion: MACS separation adult blood CD4 CD25 T cells are viable , separation of high purity . The use of IL-2 anti-CD3/CD28/beads amplified CD4 CD25 T cells can achieve significant amplification effect .
