Abstract


ObjectiveThe first experiment:to evaluate the effect of pu-erh tea (PT) on the proliferation, colon, apoptosis and necrosis of rat breast cancer line SHZ-88 cells.The second experiment:to observe the effect of PT on gastric lesion induced by N-methyl-N’-nitro-N-nitrosoguanidine (MNNG) in wistar rats.The third experiment:to observe the protection of PT on radiation damage in Kunming mice.MethodsThe first experiment:SHZ-88 cells were cultured with 5%,10%,20%PT containing serum, the ratio of inhibition of proliferation, colony formation and apoptosis was identified by MTT assay, colony formation assay and flow cytometry, respectively.The second experiment:Gastric lesion in wistar rats was induced by MNNG; the rats drink 1% PT (in distilled water) freely. Afer 22 weeks, all the rats were sacrificed to routine histopathological examination.The third experiment:The Kunming mice were given 1% or 2% PT (in distilled water) freely for a month, and then received 4Gy total body irradiation. Blood count, Blood biochemistry of LDH, ALT and AST, coefficient of liver, spleen and thymus were detected, respectively. MDA and T-SOD concentration of testis were also detected and histopathological change was observed. ResultsThe first experiment:Morphology showed the SHZ-88 cells altered to round and float. MTT result showed that PT significantly inhibited the proliferation of SHZ-88 cells with time-and dose-dependent. Colony formation assay revealed that PT obviously decreased the the colony formation of SHZ-88 cells. Flow cytometry showed that PT significantly induced the apoptosis and necrosis of SHZ-88 cells.The second experiment:Visual observations showed gastric wall in model group became thickened, increased fragility and lost its elasticity. HE stains showed the incidence of tumor, hyperplasia, edema, keratinization and inflammation in PT intervention group were significantly lower than that of in model group.The third experiment:The results showed that PT reduced the serum LDH and ALT evel, the expression of MDA and increased the expression of T-SOD in testis with dose-dependent.ConclusionsPT containing serum can inhibit the proferliration and conlon of rat breast cancer line SHZ-88 cells and induce the apoptosis and necrosis. PT can protect the rat stomach mucosa lesion induced by MNNG. Pu-erh tea can also lighten the radiation damage of mice.
