Abstract


The combination of nano-materials and bio-medicine in the development ofdrugs, in terms of disease prevention and treatment are of great significance.Layer-by-layer self-assembly (LBL) as a classical method for the preparation ofnanomaterials, in view of its simple operation, rich in material selection, thethickness of product can be controlled other advantages.this method will be used forthe assembly of nano-tubes and of microcapsules. In this study, preparation ofdifferent types and different materials microcapsules and make microcapsulesco-cultured with cells,research the safety and feasibility of microcapsules as adrug carrier. The main contents are as follow:Fabrication of multicomponent microcapsules via electrostatic interaction.Respectively with manganese carbonate and silicon dioxide microspheres astemplate, electrostatic interaction as a driving force, through layer-by-layerself-assembly methods prepared different types of microcapsules, such as withchitosan (CHI) and alginate(ALG) as materials to assemble biocompatiblemicrocapsules, with polystyrene sulfonate (PSS) and polyallylamine hydrochloride(PAH) as materials to assemble polyelectrolyte microcapsules, And through thepreparation of fluorescent markers with fluorescence properties of microcapsules.Co-cultured of different types of microcapsules with cell.Through differenttypes different outer and different forms of microcapsules co-cultured with cells,observing and analyzing the capsule type, surface electric charge, shape and otherfactors on the impact of phagocytosis. Human cervical carcinoma cells (Hela) havemany characteristics,which can be continuous passage, divide endlessly withoutdying,rapid proliferation and highly infectious, therefore, the experimental section,select human cervical carcinoma cells (Hela) as co-cultured cells in the experiment.Determining types of capsules as a drug carrier and to achieve the release of thedrug package. Observing the condition of capsules and cells through CLSM, basedon the results of experiments, which capsules with obvious phagocytosis can beselected as a drug carrier, the anti-cancer drugs assembled with the selected capsule,to achieve the preparation of the drug carrier. Finally, by comparing the growth andapoptotic effects of cancer cells before and after co-culture and the release ofanticancer drugs to study the selection and application of capsules as a drug carrier.
