Objective: To investigate the estrogen receptor-related receptor ERR (estrogen receptor related receptors ERR) α, β, γ relationship between VD patients peripheral blood mononuclear cell expression levels of estrogen efficacy. Method: to estrogen therapy 80 cases of post-menopausal weight degree VD patients (effective group of 40 cases, invalid group of 40 cases) and the purposes of estrogen therapy before and after the blood sample as the object of study, with the age of the 80 cases of normal elderly post-menopausal women are using to as controlled study. VD using the Mini Mental State Examination (MMSE-R) and the Clinical Dementia Rating Table judgment. Normal postmenopausal women serum estradiol concentration was used as control and analysis and related complications between groups of patients before and after treatment, serum estradiol concentrations were measured by ELISA. By semi-quantitative RT-PCR was used to detect the the VD effective group and ineffective group of patients ERRs family subtype mRNA expression before and after treatment in estrogen isoforms of the normal control group to contrast; using Western Blot blotting detection The ERRs various subtypes protein expression before and after treatment of estrogen. Results: 1. Elderly women with vascular dementia in patients with serum estradiol levels significantly lower than the age and sex-matched normal population, estrogen replacement therapy in serum estradiol levels. Vascular dementia patients with serum estradiol levels and patient at the same time suffering from diabetes, diabetic patients with serum estradiol levels before and after treatment were significantly lower than those without diabetes. 3.RT-PCR and Western Blot Western blot: ERRα, β and γ mRNA and protein in patients with VD special mode of expression: ERRα low expression in ineffective group, effective group highly expressed, but the expression below normal control; ERRβ basically unchanged between effective group and ineffective group and the normal control; ERRγ high expression in ineffective group, low expression in the effective group, the normal control was the lowest. Clinical treatment of severe VD patients can be detected in peripheral blood mononuclear cells ERRα / ERRγmRNA or protein expression ratio speculated that the treatment effect, or as severe as estrogen therapy in patients with VD indications. Conclusion: Compared with normal controls, patients with vascular dementia, peripheral blood mononuclear cells ERRα expression was lower, higher ERRγ expression. Estrogen therapy patients, ERRα expression was significantly increased ERRγ expression was significantly decreased. This study suggests that ERRα / ERRγ change in the ratio may be related to the incidence of vascular dementia, the efficacy of estrogen close relationship, this ratio can be a clinical indicator of severe estrogen therapy in patients with VD. The EERs the molecular basis of estrogen treatment of severe vascular dementia.
