Effect of Neoadjuvant Chemotherapy on Micrometastasis in Breast Cancers’ Peripheral Blood by Means of Detection of hMAM RNA

OBJECTIVE:To observe the change of circulating tumor cells in blood of the patients with breast cancer before and after neoadjuvant chemotheraphy and discuss its significance.METHODS: The expression of hMAM mRNA of twenty-three breast cancer cases were detected by Nested RT-PCR.All cases underwent TA chemotheraphy and then the expression of hMAM was detected again.The patients’reactions to the chemotheraphy were also observed.RESULTS: The hMAM expression is significant corelated with clinical stage and lymph node state,but is no significant association with ER,PR,Cerb-2 expression status or pathological type . Sixteen cases(69.6%) have a positive expression of hMAM in peripheral blood.After neoadjuvant chemotheraphy,seven case achieved PR and nine cases achieved SD,no CR or PD cases.The hMAM expression became negative in three patients and declined in all PR cases,the difference was statistic significant,P=0.009. Four patients had a declined expression of hMAM in SD group,but the difference wasn’t statistic significant,P=0.074.The declination of hMAM expression was closely correlated with patients’reaction of neoadjuvant chemotheraphy,r=0.928.CONCLUSION:Neoadjuvant chemotheraphy can reduce the circulating tumor cells in blood of the paients with breast cancer and the reduction of circulating tumor cells may be a early predictior of neoadjuvant chemotheraphy.
