Abstract


The related bacteria diversity of Populus euphratica, Haloxylon ammodendron,Phragmites australis and Tamarix ramosissima Ledeb, Which are the dominant plants in theKumtagh Desert, China, were studied.Traditionally microbiological isolating culture andmolecular biology technologies16S rDNA sequence analysis have been used to study diversityof polpulation quantity and community structure of of these plants roots and three organs ofHaloxylon ammodendron..The bacteria in the roots of different plant samples were isolated by two cultue medium,NA and R2A. There are210strains of bacteria were isolated in this experiment were groupedinto36know genera.54strains of bacteria were isolated from Phragmites australis fromXiaoquangou spring were grouped into16known genera.37strains of bacteria were isolatedfrom Populus euphratic from Xiaoquangou spring were grouped into12known genera.47strains of bacteria were isolated from Tamarix ramosissima from Shule river were groupedinto12known genera.29strains of bacteria were isolated from Tamarix ramosissima fromAqike valley were grouped into7known genera.43strains of bacteria were isolated fromHaloxylon ammodendron from the opposite of Mingsha mountain were grouped into11knowngenera. Otherwise, On the other side, there are big different in isolated bacteria spaciesbetween two kinds of Tamarix ramosissima Ledeb collected from different sample site,Which can be speculated that differernt related bacteria community structure for same plant indifferent place. NA and R2A are complementary in isolate bacteria species.The bacteria in roots, stems and leaves of Haloxylon ammodendron were isolated by NAand R2A culture medium. Stem samples have the highest number of bacteria (1.25×104cfu/gfw), roots had the middle values (1.11×104cfu/g fw), whereas leaves had the lowest values(0.09×104cfu/g fw), there is a big gap between leaf compare to stem and root. A total of89bacteria strains isolated from three organs of Haloxylon ammodendron were grouped into18known genera.43strains isolated from the roots were grouped into11known genera;20strains isolated from the stem were grouped into9known genera;26strains isolated from the leaves were grouped into8known genera, and there was no obvious dominant genera. Theresults suggested that bacteria have the organ specificity. Our results revealed Part of generahave showed organ specifity.There are256strains of bacteria were isolated in this experiment were grouped into39know genera.25sequencing strains may be potential new bacteria,9.76%of the total.
