Abstract


Dendrobium denneanum is the stem of Orchidaceae plant Dendrobium denneanum Kerr.,it possess effects which is replenishing the stomach and promoting the production of the body fluid, nourishing yin and clearing away heat from the lung,for Ranging jin kui, dry mouth vexed thirsty, eat less retching, virtual hot after illness, eyes dark unknown disease and syndrome, etc. This topic based on the national science and technology support program" Dendrobii Caulis standardized planting and its SOP with optimization upgrade research", studied on the quality standard of Dendrobium denneanum.We studied on the qualitative identification and inspection research, using microscopic identification method to indentify powder and slices. Using the TLC identification control medicinal materials; referencing to the Chinese pharmacopoeia (2010) edition under items,we test the moisture,total ash,acid insoluble ash, Heavy metal and arsenic salt. We measured water and50%ethanol as solvent of the extract to determination.According to results of Materia Medica,we get the Strait characteristic and microscopic characteristics. The moisture was5.68%～9.85%, the total ash was1.39%～3.22%, the acid insoluble ash was0.08%-0.36%, heavy metal content is Less than10ppm and arsenic salt content is less than2ppm.,the water-soluble extraction was21.16%-33.19%, the ethanol-soluble extraction was11.39%-23.98%.So we suggest moisture should not be above11.0%, total ash should not be above5.0%, acid insoluble ash should not be above2.0%, water-soluble extraction shall Shall not be less than20.0%, ethanol-soluble extraction shall Shall not be less than10.0%.We have established a method for determination Polysaccharide and Gigantol contents in Dendrobium denneanum Kerr.. Respectively for two methods of accuracy, repeatability,precision,stability test and determination of polysaccharide in six batches Dendrobium denneanum Kerr.,the comparison showed that the better method to determine the content was improved phenol sulfuric colorimetric acid,the cotent of polysaccharide was6.15%-11.02%;To optimize the process of extracting effective constituents of gigantol from Dendrobium denneanum Kerr. by Central Composite Design-Response Surface, the cotent of gigantol was0.0041%-0.0344%. According to results of Materia Medica, the cotent of polysaccharide should be more than6.0%, the cotent of gigantol should be more than0.0040%.In order to improve Dendrobium denneanum Kerr. TLC identification,we studied on the quality of control medicinal materialsand established standard.The results of this study provide a scientific basis for Dendrobium denneanum Kerr. Quality standards to establish and improve.
