Abstract


Objective:RCC(renal cell carcinoma),kidney cancer for short,is one kindof malignant tumour come from Urinary epithelial cells of kidney.Atpresent,from2to3percent of Malignant tumor in adults is RCC.According tothe epidemiology statistics of authority body,the morbidity of RCC in ourcountry take on rising trend.LncRNA(Long noncoding RNA) is always considered by scientists to benoise gene in process of transcription, but recently scientist believe LncRNAplay a major role in a number of biological processes,such as genomicimprinting,chromatin remodeling and transcriptional activation.Theinvestigate of LncRNA has become research focus in recent years.It has beenproved out that HOTAIR is overexpressed in a variety of tumors.HOTAIRhas been supposed to be a valuable molecular tumor markers.Some studieshave confirmed the mechanism that HOTAIR serve as scaffold moleculewhich control the methylation and demethylation of EZH2and other relatedmolecules.This researchmainly test the expression of HOTAIR and EZH2inRCC and research the clinical significance.Methods:1Test the mRNA expression of HOTAIR and EZH2in cancer tissues andprecancerous lesions from fifty-eight cases of RCC.2Use statistical software SPSS17.0for statistical analysis.Result:1The mRNA expression of HOTAIR in Renal Cell CarcinomaThe relative expression quantity of HOTAIR in renal cancer tissues andnormal tissues:0.75±0.19and0.38±0.09.The mRNA expression of HOTAIRin renal cancer tissues is significantly higher than that in precancerouslesions(P<0.05). 2The mRNA expression of EZH2in Renal Cell CarcinomaThe relative expression quantity of EZH2in renal cancer tissues andnormal tissues:2.03±0.46and0.64±0.15.The expression of EZH2in renalcancer tissues is significantly higher than that in normal tissues(P<0.05).3The relationship between the expression of HOTAIR、EZH2and personaldetails like sexuality,age,smoking history,hypertension,or clinicopathologicaldata like clinical staging,lymph node metastasisThe mRNA expression of HOTAIR in renal cancer tissues which comefrom patients not less than sixty years old is0.75±0.22, and that come frompatients less than sixty years old is0.75±0.14,There is no significant differentbetween them(P>0.05).The mRNA expression of EZH2in renal cancertissues which come from patients not less than sixty years old is2.10±0.49,and that come from patients less than sixty years old is1.71±0.71, Thereis no significant different between them(P>0.05).The mRNA expression of HOTAIR in renal cancer tissues which comefrom male patients is0.73±0.20,and that come from female patients is0.78±0.17,There is no significant different between them(P>0.05).The mRNAexpression of EZH2in renal cancer tissues which come from male patients is1.98±0.48,and that come from female patients is2.18±0.40, There is nosignificant different between them(P>0.05).The mRNA expression of HOTAIR in renal cancer tissues which comefrom patients having smoking history is0.75±0.16,and that come frompatients not having smoking history is0.74±0.23,There is no significantdifferent between them(P>0.05).The mRNA expression of EZH2in renalcancer tissues which come from patients having smoking history is1.98±0.47,and that come from patients not having smoking history is2.20±0.42,There is no significant different between them(P>0.05).The mRNA expression of HOTAIR in renal cancer tissues which comefrom patients having hypertension is0.72±0.19,and that come from patientshaving hypertension is0.81±0.18,There is no significant different betweenthem(P>0.05).The mRNA expression of EZH2in renal cancer tissues which come from patients having hypertension is2.07±0.43, and that come frompatientsnot having hypertension is1.74±0.76, There is no significant differentbetween them(P>0.05).The mRNA expression of HOTAIR in renal cancer tissues which comefrom Ⅲ-stage renal cell carcinoma is0.91±0.15,which significantly higherthan that in Ⅱ-stage renal cell carcinoma(0.76±0.12)(P<0.05).The mRNAexpression of HOTAIR in renal cancer tissues which come fromⅡ-stage renalcell carcinoma is significantly higher than that in Ⅰ-stage renal cellcarcinoma(0.58±0.07)(P<0.05).The mRNA expression of HOTAIR in renal cancer tissues which havinglymph node metastasis is0.93±0.15,which significantly higher than that nothaving lymph node metastasis(0.65±0.13)(P<0.05).The mRNA expressionof EZH2in renal cancer tissues which having lymph node metastasis is2.40±0.44,which significantly higher than that not having lymph nodemetastasis(1.84±0.34)(P>0.05).4The correlation analysis between the mRNA expression of HOTAIR in renalcancer tissues and that of EZH2in renal cancer tissuesThe mRNA expression of HOTAIR in renal cancer tissues(0.75±0.19)have positive correlation with that of EZH2(2.03±0.46).Conclusion:1The mRNA expression of HOTAIR and EZH2in renal cancer tissues issignificantly higher than that in normal tissues,so the HOTAIR gene and theEZH2gene may have relationship with the occurrence and development ofRCC.2The mRNA expression of HOTAIR and EZH2in renal cancer tissuesmay have no relationship with factors like sexuality,age,smokinghistory,hypertension,and have relationship with clinical staging,lymph nodemetastasis,which may show that both of them have relationship withinvasiveness and prognosis of the RCC.3The mRNA expression of HOTAIR in renal cancer tissues have positivecorrelation with that of EZH2,which show that both of them have relationship in biological mechanisms.The combined tests of EZH2and HOTAIR mayhave reference value to judge the degree of malignancy、invasiveness andprognosis of renal cancer.
