Imaging agent is the key to both positron emission tomography (PET) and nuclear medicine. Conventional synthetic instruments for PET imaging agent production have many shortcomings:the apparatuses are very large and generally expensive.the dose for each synthesis is redundance, resulting in lower reaction rate and reaction efficiency. The purification steps are very complex.The reactions are time-consuming and difficult to achieve "on time" and "on demand" production.Each PET imaging agent requires a set of special equipment and is difficult for function extensions,which restricts the research and development of new imaging agents.Microfluidics-based PET imaging agent synthesis technologies have significant advantages. First, the imaging agents can be synthesized in a very small reaction volume,therefore have relatively high concentration of reactants and reaction rate, which can greatly reduce the amount of the substrate and reduce the difficulty of purification. Second, it can greatly shorten the synthesis time,realizing the on demand production. Third,it can significantly improve the radiochemical yield. Fourth, the reaction system is easy for safety protection.In addition, the reaction chip can be functional expanded,thus meeting the research needs. These advantages provide a new experimental platform for fast and efficient preparation of short half-life11C,13N,18F radiolabeled imaging agent.It has broad application prospects in life sciences and clinical medicine. Therefore, the study of Microfluidics-based PET imaging agent synthesis technologies is of great importance.In chapter1of this essay,the research progress of microfluidics-based PET imaging agent synthesis technologies were introduced,focusing on the synthesis of most commonly used imaging agent [18F]FDG and divided the existing synthesis system into continuous-flow systems and batch-based reactor systems, and the comparison between these two types was studied. In chapter2, a dehydration microfluidic chip for the dehydration step of the [18F]FDG synthesis process was developed. The chip is made of glass,PDMS and PC materials.A fluid half assembly and a gas half assembly were respectively formed using the irreversible sealing method.Then relying on external force,the fluid half and gas half were then reversibly bonded,forming the full microchip integrated with the glass-PDMS...PDMS-PC four layer structure.The study with this chip on the water, a mixed solution of acetonitrile and water as well as actual samples obtained satisfactory results,demonstrating the usefulness of this microchip.
