Abstract


Taxanes Taxol is a new class of effective natural anticancer drugs , sources of scarcity , high prices . The yew also known as yew , yew , in Taxaceae genus , containing the efficient anticancer ingredient paclitaxel by deforestation , endangered national key protected plants . Looking for a more efficient and cheap source of paclitaxel to the face of the growing demand for replacing yew plants , fungi , and a rich source of taxol obtained by microbial fermentation pathway is a huge space for development direction . To carry out a study of the endophytic fungi separation , screening , and the product of anti-cancer properties , aimed at finding a wider source of taxol to protect endangered yew resources , effective target new strains produce taxane substances of Taxus chinensis var. , at the same time lay the research foundation to develop new natural medicine , in order to solve some of the key issues in the paclitaxel production and final production of the drug through microbial engineering . Separation from the Southern yew bark for more than 10 years old yew endophytic fungi and 20 strains . Endophytic fungus metabolites paclitaxel (Taxol) as a standard , the use of thin-layer chromatography method ( TLC ) and high performance liquid chromatography (HPLC) separation and analysis , from screening to a Penicillium H ⑥ .1 may the ability to produce taxol compounds . By MTT colorimetric assay H ⑥ .1 fermentation product of anti-tumor properties of malignant melanoma cells A375 . Fermentation products in a concentration - dependent inhibition rate of tumor cells , the concentration of 100μg/ml , cell proliferation inhibition rate reached 50% . Initially conclusive evidence of the the cytotoxic endophytic fungi of Taxus chinensis product . A compound isolated from the metabolites of this strain , using the infrared absorption spectroscopy ( IR ) , mass spectrometry (MS), nuclear magnetic resonance (1H-NMR, 13C-NMR) as well as with the known compounds of the literature data comparison identified this compound as A tremor the toxins Penitrem A .
