Abstract


Lyme disease is a the media tick-borne Borrelia sparse the spirochete infection caused by a natural foci of zoonotic bacterial diseases, the disease is damage to multiple tissues and organs of humans and animals. Experienced Lyme disease course of the disease in humans and animals infected with Borrelia burgdorferi, showing different clinical symptoms. Lyme disease as the most common medium for one of the spread of zoonotic disease is widely distributed throughout the world, poses a severe challenge to the safety of the public health, the harm has been growing concern by the people. Domestic research on the disease is relatively small, based on isolates of Borrelia burgdorferi in different countries and regions more significant difference the Borrelia burgdorferi some of the features of the research for the disease preliminary molecular and serological detection methods research. Reference in GenBank has published the Borrelia burgdorferi gene sequence, based on 16S rRNA gene sequence design universal primers material detection of Borrelia sparse spiral body of the LAMP method; with Chlamydia, Mycoplasma and Rickettsia times's body, Babesia, Taylor no cross-reactivity between insects and other common tick-borne pathogens. Comparison with the reference PCR method, the LAMP method has good sensitivity, the detection sensitivity of the PCR method reference of narrowly Borrelia burgdorferi, A burgdorferi garinii Borrelia body DNA were 1pg 1pg and 0.1 pg; LAMP method were 0.2pg, 0.2pg and 0.02pg. Application of the established PCR and LAMP method to detect genome of 110 field collected ticks PCR detection rate of 15.5%, the LAMP detection rate of 32.7%. Application establish the LAMP method to detect different tick species collected from some regions of China, the results showed that the Dermacentor Borrelia Borrelia body infection rate was 28.6%, the Haemaphysalis the infection rate was 36.5%, the rate of infection of cattle ticks 61.0 percent, the Rhipicephalus infection rate was 54.9%. Domestic major epidemic of Borrelia sparse spiral body strains the three genotypes (narrowly defined Beth's sparse spiral body, A's sparse spiral body gamma's sparse spiral body), based on the OspA gene design specific primers material to establish can be genetic typing LAMP method; specificity analysis showed no cross-reactivity, its interspecific no cross-reactivity between the spread of the pathogen and Chlamydia, Mycoplasma and Rickettsia, Babesia, Theileria common ticks. The sensitivity analysis found that narrow Borrelia burgdorferi, A burgdorferi garinii Borrelia body-specific primers amplified its corresponding strain sensitivity 2ng, 0.2ng and 0.02ng. The results showed that the LAMP typing methods have better specificity can be used to our popular narrow Borrelia burgdorferi, A burgdorferi, and garinii Borrelia body 3 genotype strain typing identified, but this the sensitivity of the method is relatively low, is not suitable for the detection of the field samples. Cultured in vitro in the narrow sense of Borrelia burgdorferi B31 strain, preparation of whole cell antigen, sheep Lyme disease serological testing indirect ELISA, the method positive rate of Lyme disease and negative coincidence rate can reach 90.1% and 90%, and sheep mycoplasma, between the pathogens Brucella, Toxoplasma gondii, lack plastids, Babesia, Theileria positive serum did not cross-react. Application of the Gannan region of Gansu Province, the acquisition of 450 sheep serum testing, results showed: Xiahe counties, Lintan and Zhuoni,, County sheep Lyme disease infection rate was 3.3%, 6.1% and 4.1%, with an average infection rate of 5.1%, to infer that the region may be the natural foci of Lyme disease.
