Abstract


Objective: To investigate the estrogen receptor subtypes, activator protein 1 (AP-1) expression and correlation in adenomyosis (AM). Method: 1, collected 40 cases of AM ectopic endometrium of patients and their reign of endometrial tissue (proliferative phase in 22 cases, 18 cases of secretory phase), and 30 cases of normal endometrial tissue (proliferative phase of the 17 cases, 13 cases of secretory phase) using immunohistochemical methods to detect these organizations subtypes of estrogen receptor α (ERα), estrogen receptor subtype β (ERβ) and AP-1 components of the expression of c-jun. Were selected from the above-mentioned organizations ectopic endometrium and eutopic endometrium 20 cases (proliferative and secretory phase of the 10 cases) and 20 cases of normal endometrium (proliferative and secretory phase of the 10 cases), the use of Western-blot method to detect the organizations subtypes of estrogen receptor α (ERα), estrogen receptor subtype β (ERβ) and AP-1 components of the protein expression of c-jun. Results: 1 immunohistochemical results: (1) ectopic endometrium with normal endometrium: ERβ, c-jun expression histochemical score (H-SCORE) were (3.4080 ± 0.2095) min, (3.2040 ± [H-SCORE (3.7238 ± 0.1628) points above differences were statistically significant (P lt; 0.01)] and ERα / ERβ ratio (1.3411 ± 0.0824), 1.0016 ± 0.0603) lower than normal endometrium group ERα expression. (2) eutopic endometrium and normal endometrium: ERβ, c-jun expression [H-SCORE, (3.3943 ± 0.1976), (3.1775 ± 0.3090) points] higher than normal endometrium [H- ,H-SCORE (3.7238 ± 0.1628) min] and ERα / ERβ ratio (1.3411 ± 0.0824), the differences were statistically significant (P lt; 0.01). (3) ectopic endometrium group the eutopic endometrium comparison: the ERα, of ERβ, c-jun expression and ERα / ERβ ratio difference was not statistically significant (P: 0.512,0.403,0.570,0.348). (4) ERα / ERβ ratio and c-jun expression in each group: ectopic endometrium and eutopic endometrium group was negatively correlated (respectively: r = -0.683, P lt; 0.01; r = - 0.692, P lt; 0.01), normal endometrium group, no statistically significant correlation (r = -0.170, P = 0.369). 2 Western-blot results: (1) ectopic endometrium group compared with normal endometrium: ERβ, c-jun expression intensity (IOD values ??were 529.5720 ± 5.5412, 518.5390 ± 13.7481) higher than normal endometrium (IOD value were lower than normal endometrium group 352.6030 ± 31.3287,360.0030 ± 9.5776), ERα expression (IOD value of 535.2980 ± 8.4447) and ERα / ERβ ratio (1.0109 ± 0.0176) ERα expression (IOD value of 751.2165 ± 31.9425) and ERα / ERβ ratio (2.1392 ± 0.1071), the above differences were statistically significant (P lt; 0.01). (2) the eutopic endometrium normal endometrium comparison: ERβ, c-jun expression intensity (IOD value of 528.3865 ± 4.4195 515.9735 ± 11.2767) above normal endometrium (IOD value for 352.6030 ± 31.3287 360.0030 ± 9.5776), ERα expression (IOD 534.2340 ± 10.0341) and ERα / ERβ ratio (1.0125 ± 0.0171) lower than normal endometrium ERα expression (IOD value of 751.2165 ± 31.9425) and ERα / ERβ ratio (2.1392 ± 0.1071), The above differences were statistically significant (P lt; 0.01). (3) ectopic endometrium group the eutopic endometrium comparison: the ERα, of ERβ, c-jun expression and ERα / ERβ ratio difference was not statistically significant (P values ??were: 0.175,0.056,0.068,0.676). (4) of ERα / ERβ ratio and c-jun expression in each group: ectopic endometrium and eutopic endometrium group showed a significant negative correlation (respectively: r = -0.725, P lt; 0.01 ; r = -0.679, P lt; 0.01), normal endometrium correlation was statistically significant (r = -0.258, p = 0.273). Conclusion: Compared with normal endometrium AM ectopic endometrium and eutopic endometrium, ERα / ERβ ratio decreased, ERβ expression dominant, ERα expression is limited, ERβ may be from the AM the development of major role. 2, compared with the normal endometrium AM ectopic endometrium and eutopic endometrium, AP-1 expression enhanced the AP-1 and AM development have a close relationship. 3, compared with the normal endometrium AM ectopic endometrium and eutopic endometrium, ERα / ERβ ratio associated with AP-1 expression was significantly negative, ERβ on AP-1 activation and regulation may play major role.
