Background : Glucose is one of the important factors for pancreatic ductal stem cell differentiation , differentiation of insulin -secreting cells quantity and secretory capacity . Objective: To compare the pancreatic ductal stem cells differentiated cells under different concentrations of glucose -induced insulin secretion capacity , in order to determine the ideal concentration range , glucose induced pancreatic ductal stem cell differentiation and the development of standardized experimental program . Design: Randomized controlled observation . Unit: Qingdao Municipal Hospital the cell and Pancreatic laboratory material : pancreatic ductal stem cells taken from Wistar rats , met the animal ethical standards for the disposal of animals in the experiment . Methods: collagenase V digested pancreatic tissue obtained in animal experiments , dextran of Ficoll - 400 formulated as a concentration gradient , the separation and purification of the epithelial cells of the pancreas , so as to obtain the stem cells of the pancreatic duct . To obtain the stem cells were divided into 10 groups . Each group were cultured in vitro , formed through the proliferation and differentiation of insulin-secreting cells . Differentiation stage of each group are in the medium containing 10mM nicotinamide and different concentrations of glucose , the glucose concentration of respectively 5.6 mm , 10.6mm , 15.6 mM , 20.6mm , 25.6MM , 30.6mm , 35.6mm , 45.6mm , 55.6mM 105.6mM. Identification of pancreatic ductal stem cells using immunofluorescence staining . Use photochemical chemiluminescence detection of insulin secretion differentiate into insulin-secreting cells . Results: glucose concentration of 20.6mM , 25.6mM , 30.6mM group of stimulation index higher than the other groups ( P lt ; 0.05) , but three sets of pairwise comparisons showed no statistical significance ( P gt ; 0.05 ) . Glucose concentration of 15.6 mM 20.6mM , 25.6mM group of insulin secretion is higher than the other groups ( P lt ; 0.05) , but three sets of pairwise comparisons showed no statistical significance ( P gt ; 0.05 ) . Conclusion : pancreatic ductal stem cells to differentiate into insulin secretion in cell experiments , when the culture medium contained glucose concentration in the 20.6mM 25.6mN between the resulting cell insulin secretion strongest differentiation .
