Abstract


Objective: To establish experimental colored rabbit choroidal neovascularization (CNV) in animal models to study the grub extract on angiopoietin- 1 (Angl) and pigment epithelium-derived factor (PEDF) expression , so as to explore its choroidal neovascularization inhibition. Methods: 40 rabbits only color randomly divided into five groups, namely : A group : healthy control group ; B groups: model group ; C Group: Vitamin E group ; D Group: King in Pill group ; E Group: grub extract group . N = 8 ( 16 eyes ) , established by the argon laser photocoagulation way CNV model . Laser photocoagulation 24h, 7,14,21,28 d fundus color photography , 7,14,21,28 d line fundus fluorescein angiography (FFA), 14,28 d -line optical coherence tomography (OCT). Then each group of rabbits were randomly divided into two tranches, at 14,28 d were killed by air embolism and removal of posterior segment eye tissues slices hematoxylin - eosin (HE) staining , light microscopy retinal pathological changes , parallel Ang1, PEDF immunohistochemistry . Dedicated computer image analysis system for analyzing the experimental section , the statistical analysis . Results : 1.B, C, D, E group successfully established CNV rabbit model . (2) Light microscopy showed that , B, C group photocoagulation area RPE and Bruch's membrane rupture, choroidal capillary damage , fibrovascular proliferation significantly , resulting in a large number of new blood vessels . D, E groups extent of tissue damage than B, C group light , angiogenesis significantly higher than B, C group small. 3. Ang1 content expressed determination show , A group representing B, C, D, E were lower (P lt; 0.05), B, C, D, E group D group and E group than in the other groups were significantly lower (P lt; 0.05), E group than in the group D is low, but no significant difference , E groups 28 days compared to 14 days was significantly higher (P lt; 0.05). 4. PEDF expression tests showed that the content , A group than B, C, D, E group higher (P lt; 0.05), B, C, D, E group D and group E group and the other group was significantly increased ( P lt; 0.05), E group than in the D group , but the difference was not significant , E groups 28 days compared to 14 days was significantly lower (P lt; 0.05). Conclusion: grub extract can effectively inhibit experimental CNV in pigmented rabbits Angl high expression , while reducing the presence of PEDF intervention to protect the retinal tissue of CNV was inhibited .
