Abstract


Objective:To investigate the effects of GuBenKeLi on the Matrix Metalloprteinase-9 (MMP-9) and its specific inhibition---Tissue Inhibitor of Metalloprotease-1 (TIMP-1) in Lung Tissue of COPD Model Rats at both gene and protein level and clarify the mechanism of action on curing COPD to provide experimental evidences for the clinical applications with GuBenKei Capsule.Method:50 male Wistar Rats were divided randomly into five groups:Normal control group, Model control group, Prednisone group, GuBenKeLi (low dose) group, and GuBenKeLi (high dose) group, with 10 rats in each group, respectively. The COPD Model is copied in which the rats are smoked and inhaled with LPS. Meanwhile, the rats in each group were intervened with corresponding drugs and their general conditions were observed and recorded till the end of the experiments. The protein and gene expressions of MMP-9 and TIMP-1 in the lung tissue are detected using immunohistochemical method and PT-PCR.Results:Changes of bronchial and pulmonary histopathology in each group were typical, pointed out that COPD Rats Model were successful. Compared with Normal control group, the protein and gene expressions of MMP-9 and TIMP-1 are significantly enhanced in Model control group. There are significant differences between three Treatment groups and Model control group (p<0.05). From the perspective of improving the general conditions of COPD Rats Model and imbalances of protein and mRNA between MMP-9 and TIMP-1, the effect in Prednisone group is the most significant, GuBenKeLi (high dose) group comes second, and GuBenKeLi (low dose) group is the third.Conclusions:1) The GuBenKeLi is capable of releasing clinical manifestations of COPD Model rats effectively and improving airway remodeling as well as correcting the imbalances between protease and antiprotease at both gene and protein level.2) The mentioned activities have dose-response relationship, namely, the activities increase with increasing dose.
