Lentivirus Vector-mediated Human LIGHT Gene Expression Induces Inhibition in Human Large Intestine Carcinoma in Nude Mice

Objective To study the inhibition of Lentivirus vector-mediated human LIGHT gene and LIGHT expression on the protein level.Methods Preparation of HCT116 cells were enough. And then HCT116 cells which were collected were implanted randomly into the nude mice to make sure the model of human large intestine carcinoma in nude mice successful. Then the nude mice were divided into three groups subcutaneously. The first group was Lentivirus vector with LIGHT in expression group(L-LV); the second group was Lentivirus vector group(LV) and the last group was normal sodium group (NS). The L-LV group was injected Lentivirus vector with LIGHT in expression; the LV group was injected adenovirus vector and the NS group was injected normal sodium. The volume and weight of transplanted carcinoma were observed everyday. The immunohistochemistry of LIGHT protein expression should be done.Results The mean weight of transplanted carcinoma was(1.22±0.48) g、(1.06±0.29) g and (0.55±0.16) g. There was no significant difference among the tumors’volume of the three groups before the interference. After the interference, There was significant difference between L-LV group and NS group in the volum(P>0.05). The growth rate and the weight of transplanted tumors was also different. The tumor inhibition rate of the L-LV group was 49.24% and it was higher than the other groups. There was the expression of LIGHT protein in L-LV group.Conclusion The transplanted tumors can obviously inhibited by LIGHT gene and LIGHT gene can express in transplanted tumors.
