Abstract


Objective To investigate the effect of tissue inhibitor of matrixmetalloproteinase-2 (TIMP-2) on expression of matrix metalloproteinase-2 (MMP-2) the posterior sclera of the form-deprivation myopia (FDM) guinea pig.Methods 36 three weeks of age pigmented guinea pigs were randomly assigned to four experimental groups:TIMP-2 group, empty plasmid group, saline group and self-control group (9 in each group). Right eyes of all pigmented guinea pigs were given treatment, while left eyes of TIMP-2 group were self-control group without any treatment. Another nine guinea pigs were selected as control group covering the right eyes all the time. Gene-transferred TIMP-2 plasmid, empty plasmid and saline were injected in the first three groups respectively,after form-deprivation. All the guinea pigs were sacrificed and the posterior sclera of the eyeball were taken out at 2th,7th and 14th days after drug injection. We used gelatin zymography method to measure expression of matrix metalloproteinase-2.Results 1, The expression of MMP-2 in the posterior sclera of guinea pig was reduced in TIMP-2 group as compared with its self-control group and control group, which showed significant difference (P<0.05) after drug injection at 2th,7th and 14th days. But there was no significant difference (P>0.05) between empty plasmid group, saline group and control group.2、The expression level of MMP-2 in the posterior sclera of guinea pig was reduced in TIMP-2 group began to descend from the 2th day after drug injection. The level was obviously increased at 7th and then decreased at 14th day. The expression level of MMP-2 at 2th after drug injection were significant different from that at 7th and 14th days (P<0.05). But there was no significant difference(P>0.05) between 7th and 14th days.Conclusion The expression of MMP-2 in the posterior sclera of guinea pig was reduced in TIMP-2 group changes as time prolonged after drug injection. It increased tempererily and then began to descend. It may slow down the sclera remodeling of FDM guinea pig in the early stage. but this effect weaken with time.
