Abstract


In order to analysis the manage growth and quality of flue-cured tobacco were formed in Hui Li , using semi-quantitative RT-PCR technology, gel electrophoresis technology and analytical means such as tobacco region characteristic of tobacco leaf manage at the molecular level, expounds and study the main quality of flue-cured tobacco were formed related metabolic pathways and key gene for local characteristics, and the formation of smoke quality provide theoretical foundation.This research mainly from the field of tobacco plant growth situation, leaf organization structure, key metabolic pathways gene expression, metabolic pathways changes in matter, tobacco neutral to sweet material, tobacco sensory evaluation analysis absorption, the main results are as follows: 1. The study on tobacco plant development regularity:In the early period of the growth of tobacco plant, namely the transplanting to regiment tree period, Hong Da and Yun Yan 85 grow faster, K326 grew slower than Hong Da and Yun Yan 85, KRK26 grew slowest, this about their own features, the varieties of the main fan out root, aerial parts to long-term rapid growth. The middle leaf during 15- the 40d grown in the fastest phase, the stage can be considered by the regiment tree to flourish longer growth period, tobacco leaf increases speed as follows: Hong Da >Yun Yan 85> K326 > KRK26, strains Yun Yan 85> Hong Da > K326 >KRK26, leaf number growth on Hong Da and Yun Yan 85 are the same, K326 and KRK26 are the same,KRK26 leaf number were most at later growth stage.Along with the development of leaves, the thickness of mesophyll epidermis increased; the plasmid by a small, simple lamellar structure develop into the chloroplast that have a mature, thylakoid chloroplast lamellar developed structure; both the quantity and volume of starch grain increased; the quantity and volume of osmiophilic globule increased; the shape of mitochondria and nuclear changed irregular and to degrade in the end. The number and size of starch changed, which can reflect the metabolic regularity, and the change of eosinophilic granular osmium tetroxide is also thought to be related to chemical change factors of incense. 2. Tobacco plants during development law of main material metabolic pathways:The expression of key genes in sugar metabolism were examined by the method of semi-quantitative RT-PCR, in order to prove the regularity for starch synthesis in tobacco leaf development. The results showed that: the content of starch, total sugar, reducing sugar increased gradually in development; the genes expression for SPS weakened; the genes expression for AGPase, GBSSⅠand SBE also became strongly, this result reflect that the content of starch increased.Nitrogen metabolism of nitrogen assimilation process, along with the growth of the blade increases, The gene expression for NR and GS weakened gradually, this explain the tobacco age in 10-40 days were exuberant growth stage, tobacco plants tend to mature and use nitrogen reduced gradually.With the growth and development of tobacco plant, the main metabolic substances change rules are almost the same at each varieties : the content of total sugar, reducing sugar presented concussion trend of rise; the content of total nitrogen is decreased; the content of nicotine and starch increased; the content potassium, chlorine decreased gradually with growth fell.3. The content of total neutral aroma constituents and sensory evaluation The neutral aroma constituents content amount in Hongda and Yunyan 85 is higher than KRK26, this because flue-cured tobacco neophytadiene content in Huili is higher than KRK26, the neutral aroma constituents content in K326 is lower slightly than Hongda ,Yunyan 85 and KRK26 ,mainly is the neophytadiene content in K326 is lower than former three, phenylalanine degradation products and the maillard reaction products are almost the same, the cembratriendid degradation products are not significant difference, this may be the one reason of smoke zone formed its unique aroma style.According to the sensory evaluation score,KRK26 is close full marks, quality is excellent, K326 is sent, the score of Hongda and Yunyan 85 are 85 , belong to better sensory quality.The flue-cured tobacco in Huili has better sensory quality ,this due to neutral aroma constituents content are higher , the neophytadiene content in Hongda and Yunyan 85 are higher slightly than the KRK26, the other neutral aroma constituents content are not significant difference than KRK26, these factors are listens to flue-cured tobacco good quality style formed the foundation.
