In order to explore the OT and ghrelin in mice hypothalamus distribution and its relation with obesity , this study, immunohistochemical SABC method OT and Ghrelin hypothalamic nuclei regulating food intake in normal mice and obese mice expression studied and studied the injection of exogenous oxytocin on the mouse hypothalamic ghrelin expression changes , the results for the animal energy metabolism , neuropeptide relationship between regulation and obesity mechanism of important information . The main results are as follows : 1. Oxytocin in mice under the hypothalamic arcuate nucleus , ventromedial nucleus , paraventricular nucleus ( normal mice ) were more in the dorsomedial nucleus , the suprachiasmatic nucleus , lateral hypothalamic area less distributed ; ghrelin in the arcuate nucleus , under the lateral hypothalamic area , dorsomedial nucleus , paraventricular nucleus ( obese mice ) were more , but less in the suprachiasmatic nucleus , the ventromedial nucleus distribution . Show that oxytocin and ghrelin in different nuclei expression differences in the amount and location of the nucleus feeding regulatory function coincide . 2 compared with normal mice , hypothalamic paraventricular nucleus oxytocin expression in obese mice was significantly reduced ( P lt ; 0.01 ) . Feeding nuclei in the hypothalamus of oxytocin expression and normal mice basically the same, and there are no gender differences . 3 compared with normal mice , obese mice hypothalamic paraventricular nucleus , dorsomedial nucleus and lateral hypothalamic area Ghrelin expression was significantly increased ( P lt ; 0.01 ) . To obese mice for 4 weeks intraperitoneal injection of oxytocin , hypothalamic dorsomedial nucleus , paraventricular nucleus and lateral hypothalamic area Ghrelin expression was significantly decreased in the arcuate nucleus , ventromedial nucleus and the suprachiasmatic nucleus Ghrelin the expression levels have reduced ; male obese mice hypothalamic nuclei Ghrelin expression levels of obese mice than females larger decline . Conclusion: oxytocin can make the hypothalamic paraventricular nucleus , dorsomedial nucleus , lateral hypothalamic area , arcuate nucleus , ventromedial nucleus and the suprachiasmatic nucleus of Ghrelin expression decreased oxytocin and ghrelin in the central regulation of food intake forward to a mutually antagonistic relationship . Suggesting that this is one of the ways of the oxytocin role in weight loss .
