Abstract


Rice as the most important food crops , to meet about half of the world 's food needs . Worsening food crisis in the contemporary world , research aimed at increasing rice production is particularly important , and for rice genetics research can promote rice yield , and thus help alleviate the food crisis. We sift through γ -ray mutagenesis in flower 11 (ZH11) groups , separated into four thin spike mutant decreased spikelets 1-4 (des1-4), another through the EMS mutagenesis also received a dilute spike mutant des5, the main object of study is des2. des2 main phenotypes are secondary branches and lateral spikelets reduced collateral effects on other relatively weak . Genetic analysis revealed des2 F2 generation backcross with ZH11 wild-type and mutant 3:1 segregation ratio significantly , suggesting des2 seat by a recessive gene locus control ; using des2 and indica Longtefu (Longtefu) F2 generation hybrid as a mapping population , through map-based cloning technique was first discovered in the 6th on the long arm of chromosome two SSR markers RM3827 and RM528 and des2 chain , then in this interval by comparing the database japonica and indica rice genome sequence , a potential multi- polymorphic loci developed some new molecular markers , including SSR and dCAPS, and expanded mapping population , eventually des2 be framed in two dCAPS mark OsdCAPS2 and OsdCAPS3 between the physical distance of about 28kb, and found a total of SSR markers RM20378d separation . Sequence analysis shows that the gene encoding MOC1 des2 box in place of a point mutation, resulting in premature translation termination , resulting in the loss of C-terminal protein MOC1 last five amino acids. Therefore des2 phenotype is likely caused by a mutation in MOC1 . Another rare genetic analysis showed that the mutant des5 spike is caused by a single recessive gene mutation , and des2 des5 double mutant right axillary branching and development of a significant synergistic effect . This study will provide new molecular function MOC1 insights.
