Abstract

With oil pollution is increasing, efficient pollution bioremediation of oil pollution control technology has become one of the important ways, attracted widespread attention. Oil is the saturated hydrocarbons, aromatic hydrocarbons and small amounts of asphaltenes, resins and other components of a complex mixture, and a single strain that can metabolize the oil component is usually very limited, it is difficult to completely remove the oil from the environment. Mixed flora helps to improve synergies between microbial oil degradation. Therefore, how to take advantage of the interaction between different strains, optimal combination of rational and efficient degrading bacteria is currently a research hotspot. Immobilized microorganism technology enables highly intensive microorganisms and maintain its biological activity, strengthening effect on oil degradation. In this study, the laboratory has screened alkane degrading bacteria GS3C, phenanthrene and pyrene degrading bacterium GY2B degrading bacterium (GP3A and GP3B) for domestication, the results indicate that these bacteria can carry crude oil as the sole carbon source for growth and reproduction of sound, and crude oil have a certain degradation, removal rate of 30% or more. After GC-MS analysis, GS3C substantially removing the crude oil can be straight-chain alkanes, GY2B of aromatic compounds have a good degradation, GP3A the C lt; 20 of the short-chain alkanes and aromatic compounds have a certain degradation role, GP3B for alkanes and aromatic compounds degradation in general. Build efficient use of these four strains mixed flora, filter out the optimal mixed bacteria G8, for oil removal rate reached 68.76%, compared with a single strain increased by nearly 30%. The GC-MS analysis, G8 and retain the single strain GS3C GY2B degradation characteristics, can substantially removing the crude oil alkanes, while the aromatic compounds exhibit a strong degradation, the joint action of bacteria in three enhances complete removal of crude oil capacity. Bacterial degradation of the mixed performance of the G8 found the best mix mixed bacteria G8 ratio of 1:1:1, and its optimum growth conditions were: temperature is 30 ℃, initial pH 7.0, inoculum was 4%, oil concentration of 2000 mg / L, in the best environmental conditions degrade 5d after removal rate reached 69.20% of the oil. Using rice straw and corn stalks two kinds of mixed bacteria G8 vector on the surface adsorption fixed, the results showed that the immobilized bacteria on crude oil removal than free bacteria or carrier and a mixture of good bacteria, not sterile straw removal of crude oil sterilization than good. Including straw immobilized bacteria removal rate of 90% to 95%, corn stalks immobilized bacteria removal is 95% to 98%, than the free bacteria G8 alone increased nearly 40 percent. Choose not sterilized corn stalks as the best carrier fixed mixed bacteria G8, removal of crude oil reached 98.19%, the best results.
