Abstract


Objective:Using experimental method preparation vitro intestinal flora transformation model to traditional Chinese medicine decoction of Radix glycyrrhizae through the biotransformation comparison decoction of Radix glycyrrhizae before and after the transformation main chemical composition and therapeutic effects of intestinal flora changes, analysis of biotransformation sulation into effect.Methods:(1)The rich in intestinal bacteria with fresh dejection of guizhou miniature pig supernatant and decoction of Radix glycyrrhizae in anaerobic conditions temperature hatch, preparation personification bowel flora transformation model.(2)Using HPLC method detection before and after transformation glycyrrhizic acid, glycyrrhetinic acid,liquiritin,liquiri -tgenin and other main compositions, and explore the changes of intestinal flora composition can influence more than. (3) The cup-plate and tube method, observation licorice water extraction liquid transformat before and after the in vitro antimicrobial effect change; Through of auricle tumefaction mice caused by experiment and ammonia lead cough experiment, observation licorice water extraction liquid transformation of before and after the degree of auricle tumefaction mice incubation period and cough cough, the influence of frequency conversion intestinal flora, understand the effect detoction of Radix glycyrrhizae such as anti inflammatory, antitussive effect.Results:(1)Decoction of Radix glycyrrhizae can be this experiment preparation intestinal flora model transformation. (2)The HPLC method detected glycyrrhizic acid, glycyrrhetinic acid, liquiritin,liquirtgenin into a different degree of change before. (3) Decoction of Radix glycyrrhiza on escherichia coli, staphylococcus aureus, micrococcus citreus,pseudo monas aeruginosa, bacillus subtilis all has bacteriostasis.Biotransform of decoction Radix glycyrrhizae after of staphylococcus aureus bacterio static effect and pseudomonas aeruginosa antibacterial, sterilizing effect before more obviously than transformation;In the anti-inflammatory experiments, compared with normal saline groups by intestinal flora, transformed into the liquorice water extraction liquid high, medium and low dose group of all can inhibit xylene-induced ear edema in mice, hind paw edema degrees caused a statistically significant difference (P<0.05 or P<0.01), and transformation former licorice water extraction liquid high dose group compared transformed into the middle, high dose group of difference was statistically significant (P<0.05 or P<0.01);In the antitussive experiments, and physiological saline into group compared before and post-transform high-dosage groups high, medium and low dose group of all can prolong mice cough cough and reduce the number of incubation period is statistically significant difference (P<0.05 or P<0.01), and transformation former licorice water extraction liquid high dose group compared transformed into the middle, high dose group of difference was statistically significant (P<0.05 or P<0.01).
