Study on Non-aqueous Polar Drug Carrier for Vitamin C and Its Rheological Properties

Abstract


In this paper, glycerin(GLY) containing carrageenan as a new non-aqueous carrier for vitamin C was prepared, and the non-aqueous polar media microemulsion as carrier for vitamin C was prepared by using poly(4) glycerol mono alkyl ether(PGL) as emulsifier, isopropyl myristate (IPM) as oil phase, symdiol 68(1,2-hexylenediol 65%,1,3-octylenediol 35%) as co-emulsifier. The stability of vitamin C in non-aqueous carrier and rheological properties were investigated, and the removal effect on 1,1-Diphenyl-2-picrylhydrazyl radical (DPPH·) of vitamin C in the carrier was studied. The main conclusions were made as follows:1) For the glycerin/carrageenan carrier, the retention rate of vitamin C increases with the increase of carrageenan content, and the maximum retention rate was 99.53% at room temperature. The glycerin/carrageenan carrier can protect vitamin C well. With the increases of water in the glycerin containing carrageenan formula, the retention rate decreases. The removal percentage of DPPH·increased with the increases of vitamin C, and the maximum percentage was 86.89%, showing good ability for removing radicals.2) The rheology for glycerin/carrageenan drug carrier showed that, for the glycerin/carrageenan carrier, the viscosity increased with the increases of carrageenan and possessed shear-thinning property. When the mass ratio of carrageenan to glycerin was over 0.6%, the carrier possesses thixotropic loop. With the addition of vitamin C, the viscosity decreased and the thixotropy disappeared.3) Using glycerin as non-aqueous polar media, carrageenan as thickener, the GLY/carrageenan /IPM/PGL/S68 non-aqueous microemulsion carrier for vitamin C was obtained for the first time, and the non-aqueous microemulsion carrier was Newtonian fluid. The retention rate of Vitamin C increased with the increase of carrageenan content. The maximum retention rate was 98.71% at room temperature. The retention rate was nearly the same as glycerin/carrageenan carrier, showing that the non-aqueous microemulsion carrier possessed good ability for protecting vitamin C.The study in this paper provided a good foundation of drug delivery system for Vitamin C.
