Background Gallstones are a common disease. In recent years, its incidence is increasing every year, no effective prevention method, therefore consuming a large amount of medical resources. The formation of gallstones is the the pathological mineralization process of a human body. In the process of stone formation, the structure and composition of gallstones record the stones forming all kinds of information in the process. Through the study of the structure and composition of the gallstones can provide a theoretical basis for gallstone formation mechanism and further prevention measures to find a scientific and reasonable treatment program has played an important role in help. Because of the structure and composition of gallstones mostly from the perspective of a particular subject, coupled by the experimental conditions and the reasons for the limitations of technology, the research on the analysis of the structure and composition of gallstones encountered a bottleneck problem. In recent years, with the development of science and technology, traditional research methods have been large improvements, more advanced laboratory instruments and equipment is widely used in scientific research, provide a strong support for the experimental analysis of gallstones. The same time, with a change in mindset, a growing number of disciplines and concepts cross each other, penetration and provide a good platform for the study of gallstones. The purpose of this paper applied geology, mineralogy, crystallography disciplines modern research methods and and means of detection, the micro-structure and material composition of gallstones from the point of view of the interdisciplinary preliminary analysis for research gallstone formation The process and mechanism to provide a theoretical basis. The research and application of the method of optical microscopy, new polarizing microscope and nano-scale high-resolution scanning electron microscopy to observe the microstructure of gallstones; Fourier transform infrared specular reflection spectroscopy of gallstones in the main ingredient of organic and inorganic substances positioning, qualitative and quantitative determination of gallstones in the major and minor inorganic elements; application of X-ray fluorescence spectrometry determination; application of X-ray diffraction analysis of the composition and ingredients of of gallstones crystal substances. Results 1 optical microscope, polarizing microscope, scanning electron microscope: Most cholesterol stones there is a periodic layered structure, these stones are mostly composed of irregular shapes crystal structure of the layered structure of the core and the periphery composed. These layered structure is mainly composed of densely arranged in clusters of cholesterol crystals, cholesterol crystals arranged showing some regularity, showing the rod, layered, streak, plate, radial, micaceous, wavy or irregular arrangement . Bile pigment stones not seen such a layered structure, lumps, large and small particulate matter, these particles or scattered or gathered distributed internal stones arranged in no certain regularity and particles between larger voids. The mixed stones at the same time particulate matter has a crystal structure of cholesterol stones and bile pigment stones. Black stones in the non-the obvious core area and peripheral layered structure, mainly composed of dense small particles distributed in stones internal. In addition, in the presence of a nano-particulate material is also found in the gallstones. Fourier transform IR specular reflectance spectroscopy: gallstones exist mainly cholesterol, bilirubin, calcium carbonate, calcium phosphate, calcium stearate, organic and inorganic substances; different positions of the same substance in a variety of stones different content; cholesterol and in addition to the rest of the calcium salt of bilirubin calcium content within the same stones to its location related; cholesterol content in the core and nuclear stratified structure of stones, except bilirubin calcium shell are higher than the other substances. 3.X-ray fluorescence spectrometry: essential constants of inorganic elements can be found in gallstones, most of the essential trace elements also exist in gallstones. Although the content difference between the individual elements is large, but a positive correlation between the various calculi highest Ca content, and the Ca and S, P, of Mn content. Fe, Al, Mg, Si content of between also showed a certain degree of positive correlation. 4.X-ray diffraction: crystal composed of various types of stones obvious. The mixed stone body mixture of crystalline and amorphous substances as a main component, mainly cholesterol and bilirubin. Conclusion various types of gallstones diversity microstructure. Most cholesterol calculi in the existence of a periodic layered structure, primarily by the regular arrangement of cholesterol crystals. Gallstones there still exist a nano-particulate material. Both commonness of the main ingredients of the substance, the content and distribution of various types of gallstones, and heterogeneity. And the content and distribution of gallstones with a certain regularity.
