The Influence in Different Route of Medication with Vitamin A on CD4~+CD25~+ Regulatory T Cell and Cell Factor of IL-4、 IFN-γ in a Rat Persistent Asthma Model

Abstract


Objectives Through the establishment of a rat persistent asthma model, we took different route of medication with vitamin A as the interventions, so as to comparing asthmatic rats blood CD4 ~+ CD25 ~+ regulatory T cells (CD4 ~+ CD25 ~+ Treg) and serum cytokines IL-4, IFN-γchanges, thus realizes the nosogenesis of CD4 ~+ CD25 ~+ Treg in asthmas, and also discusses CD4 ~+ CD25 ~+ Treg and serum cytokines IL-4, IFN-γchanges in asthmatic rats blood ,which given the different route of medication with vitamin A. So finding the theory and provide basis for vitamin A in the prevention and treatment of bronchial asthma.Methods First at random 60 adult female SD rats divide into two groups, one is Group A, the group of asthma, 28 adult female SD rats, the other is Group B, the common group, 28 adult female SD rats. The rats of Group A were injected into abdominal cavity with sensitization fluid of 1.0 ml / only / times (0.9% normal saline) on the 14th days of the experiments, rats had been inhaled 0.9% normal saline solution for 21days. After three weeks, 32 rats were divided into A1, A2, A3, A4 four groups, A1 group is taking vitamin A, A2 group is weeping vitamin A in nose, A3 group is taking edible oil, A4 group is weeping edible oil in nose. On the 1st and 7th days of experiments: rats of Group B were injected into abdominal cavity with sensitization fluid of 1.0 ml / only / times (the mixtures of 0.08% OVA and aluminum liquid). On the 14th days of the experiments, rats had been inhaled 1% OVA solution for 21days in order to establish persistent asthma model , daily one time, 30 minutes each time. After three weeks, 32 rats were divided into B1, B2, B3, B4 four groups, B1 group is taking vitamin A, B2 group is dropping vitamin A in nose, B3 group is taking edible oil, B4 group is weeping edible oil in nose. After stimulation 4th week and the 5th week, peripheral venous blood from the orbital venous plexus were 50μl, to flow cytometry to detect CD4~+CD25~+Treg and determinate IL-4, IFN-γby the ELISA law.Results (1) CD4 ~+ CD25 ~+ Treg results: in intervene 3rd week, common group CD4~+CD25~+Treg compared with asthma group is distinct advanced, by analysis of statistics, there has remarkable significant difference. In intervene the 4th week, the result of CD4~+CD25~+Treg in taking and dropping nose vitamin A compared with taking and dropping nose edible oil, which was slowly heighten, by analysis of statistics, there has no significant difference. The result of CD4~+CD25~+Treg in taking vitamin A compared with vitamin A dropping nose , which was slowly heighten, by analysis of statistics, there has no significant difference. In intervene the 5th week, the result of CD4~+CD25~+Treg in taking and dropping nose vitamin A compared with taking and dropping nose edible oil, which was clear heighten, by analysis of statistics, there has obvious significant difference. The result of CD4~+CD25~+Treg in taking vitamin A compared with vitamin A dropping nose , which was slowly heighten, by analysis of statistics, there has obviously significant difference.(2) cytokines results: in intervene 3rd week, the results of IL-4 and IFN-γin common group compared with asthma group is distinct advanced, by analysis of statistics, there has remarkable significant difference. In intervene the 4th week, in asthma group, the result of IL-4, IFN-γin taking and dropping nose vitamin A compared with taking and dropping nose edible oil, which has slowly differences, by analysis of statistics, there has no significant difference. The result of IL-4, IFN-γin taking vitamin A compared with dropping nose vitamin A, IL-4 was slowly heighten, by analysis of statistics, there has no significant difference, and IFN-γwas slowly drop, by analysis of statistics, there has no significant difference. In intervene the 5th week, the result of IL-4, IFN-γin taking and dropping nose vitamin A compared with taking and dropping nose edible oil, which has obviously differences, by analysis of statistics, there has clearly significant difference. The result of IL-4, IFN-γin taking vitamin A and dropping nose vitamin A, IL-4 was heighten, by analysis of statistics, there has clear significant difference, and IFN-γwas drop, by analysis of statistics, there has no significant difference.Conclusions (1) There is a misbalance CD4~+CD25~+Treg in asthmatic rats ,and clear drop with common rats. (2) the results of IL-4 in asthmatic rats is higher than common rats; the results of IFN-γin asthmatic rats is lower than common rats. (3) drop nose and take vitamin A course the results of CD4~+CD25~+Treg slowly higher. (4) drop nose and take vitamin A course the results of IL-4 slowly down, and the results of IFN-γslowly high. (5) the results of CD4~+CD25~+Treg in dropping nose with vitamin A is higher than taking vitamin A. (6) the results of IL-4 in dropping nose with vitamin A is lower than taking vitamin A; the results of IFN-γin dropping nose with vitamin A is higher. than taking vitamin A.
