Abstract


The European eel is our main eel breeding varieties. With the rise of farmed European eel diseases also showed a tendency to spread, morbidity and mortality increased year by year, to become one of the important factors restricting the development of the European eel aquaculture. Against the European eel, a wide range of pathogens, disease prevention and treatment technology lag, especially viral diseases, the eel viral diseases is still in a relatively superficial level. Impact eel viral disease in-depth study of the main reason is the lack of a suitable cell model. So far, not yet seen the European eel cell lines reported. In order to establish a separation of the European eel viral diseases, the identification of the cell model, in-depth investigation of the eel viral disease prevalence and spread of the experimental artificial European eel pectoral cell culture adapted strains (21 generations) as experimental material, the use of artificial medium in vitro culture methods, explore the optimum conditions of the European eel pectoral passage cells in vitro; based on optimal medium by adding a basic fibroblast growth factor (bFGF) and insulin-like growth factor 1 (IGF passaged cell -1) may promote the growth factors, and to analyze the impact of the European eel pectoral cell growth rate. By observing compare different medium (L-15, RPMI-1640, high-sugar type DMEM) serum (BS and FBS), and cultured under the conditions of the serum concentration (5%, 10%, 15%, 20%) European pectoral cells growth conditions, and within a limited time to collect the cells, cell count, draw the pectoral cell growth curve analysis of different medium and serum components and the influence of the concentration on the European eel pectoral cell growth state. Medium optimization results show that: L-15 medium best suited the European eel pectoral cell growth, the worst effect of RPMI-1640, followed by the high sugar type DMEM; FBS (fetal bovine serum) is superior to the BS (fetal calf serum) which 10? S added amount. bFGF and IGF1 add test results suggest that: 10? S L-15 full price of medium conditions, bFGF significant role in promoting European the pectoral cell growth, the best effect concentration for 10ug / L; IGF-1 (5ug / L-50ug / L) is not conducive to the European eel pectoral cell growth. Above study in the three media tested, two kinds of serum and two kinds of cells and promote the growth factor screened pectoral fin cells in Europe optimum medium formula of the full price, while initially confirmed bFGF promote long European pectoral cell , while adding IGF-1 is not conducive to the pectoral cell growth in Europe. The results shorten the European eel pectoral cell doubling time, improve pectoral fin cell culture cycle is limited biomass provides a practical, effective formulation and training methods, this method eel virus venereal original separation, identification and vaccine preparation has potential application.
