Abstract


Objective of hTERC FISH detection of cervical tissue HER-2 gene expression, to analyze the relationship with clinicopathological parameters, explore of hTERC, HER-2 in cervical cancer, the possible role of the development. Were collected from March 2008 to March 2009 in Qingdao University Medical College, subsidiary hospital gynecological treatment of 162 cases of patients of cervical exfoliated cell samples, of which 45 cases of normal and inflammation by 29 cases of CIN I who, CIN II - III by 38 cases of 50 cases of cervical cancer. Collected over the same period of surgical resection or biopsy of cervical tissue specimens of 63 cases, in which the pathological findings confirmed normal and inflammation in 13 cases, 12 cases of CIN I by CIN Ⅱ - Ⅲ 20 cases of cervical cancer in 18 cases. Five producer methods, fluorescence in situ hybridization (FISH) detection of cells and tissues collected hTERC and HER-2 gene expression and compare the difference between the five producers methods. Results for the detection of 225 specimens, including 20 cases of direct smear method in 12 cases, 60 cases of saline law, hypotonic producer, TCT producer method in 70 cases, paraffin-embedded tissue sections Paraffin 63 cases, with a total the hybrid success rate of 76.4%; TCT Paraffin, paraffin-embedded tissue sections Paraffin hybridization success rate with other producers Law hybridization success rate difference was statistically significant (P lt; 0.05), which TCT producer The method of hybrid highest success rate to 87.1% of hTERC, HER-2 gene expression in 40 cases of normal cervical, and other disease group were expressed. CIN Ⅰ group, gene hTERC CIN Ⅱ - Ⅲ group, the positive expression rate of cervical cancer group gradually increased, the difference was statistically significant (P lt; 0.05); gene HER-2 in the group of CIN I, CIN Ⅱ - Ⅲ group, cervical cancer group The positive expression rates were 6.5%, 28.6%, 59.3%, followed by gradually increased, the difference was statistically significant (P lt; 0.05) of hTERC, HER-2 gene expression and the high histological grade were significantly related (P lt ; 0.05). of hTERC HER-2 gene expression in cervical lymph node metastasis and degree of differentiation were significantly associated (P lt; 0.05), with age, tumor size, clinical stage, histological type and pathological type related (P gt; 0.05). Conclusions contrast five FISH producer, the detection of hTERC of cervical cancer screening, the the TCT producer method is superior to other methods, and guide the treatment of cervical lesions and cervical cancer, the actual method of paraffin-embedded tissue sections more meaningful. of hTERC, HER-2 gene expression may play an important role in cervical cancer occurrence and development of cervical lesions detected in exfoliated cells or tissues of hTERC, HER-2 gene can be used as early screening of cervical cancer, disease surveillance, assessment prognosis secondary indicators.
