Abstract

The existing studies confirm Probiotics inhibit diarrhea caused by rotavirus and HIV, but antiviral activity of probiotics is still a lack of systematic research. Therefore, the present study, the method of MTT and NDV-HA, respectively chick embryo fibroblasts (CEF) and chicken embryo intestinal tissue in vitro, a preliminary evaluation of the impact of probiotics on the the NDV the hemagglutination price and the inhibition rate, so as to explore the probiotic the antiviral activity of the bacteria and the analysis of its possible mechanisms. The first experiments with Gram stain method to detect the ability of probiotics CEF and chicken embryo intestinal tissue adhesion results determine probiotics on CEF adhesion, but significantly lower than the chick embryo intestinal tissue adhesion; cell activity after probiotic treatment was measured using the MTT assay, derived by statistical analysis of the maximum non-toxic concentration of probiotic cells to Lactobacillus casei 1011cfu/ml Lactobacillus acidophilus 1012cfu/ml, probiotic culture supernatant 1:2 and the diluted solution probiotic inactivated bacterium and crushing bacterial component were not apparent in the experimental concentration cell poison phenomenon; susceptibility testing screening of Lactobacillus casei and Lactobacillus acidophilus are highly sensitive to antibiotics - gentamicin . Then, based on the above experimental results, the probiotic cells Antiviral Research. The experiment grouped as follows: Group 1: Probiotics in advance with the two kinds of cells were incubated then challenged; Group 2: the two kinds of cells infected with the virus and then access probiotics; Group 3: probiotics with the virus at the same time access cells; 4 groups: probiotics control; Group 5: virus control; six groups: normal cell control. The results show: the in vitro probiotic the NDV hemagglutination price impact the experiment, the choice of five kinds of probiotics and their culture supernatants are very significantly reduces the NDV hemagglutination price, but exists between the various strains different degrees the differences; experiment two kinds of bacteria and their culture supernatants of NDV hemagglutination price has no effect This result suggests that probiotics may have directly undermined the role of the virus, and bacteria-specific. MTT assay cell activity, after the processing of the probiotic and NDV viral inhibition rate calculated by the formula, the results show either on CEF or in the chick intestinal tissue cells, probiotics can significantly improve the inhibition of NDV rate, and the probiotics this role has the dose-response relationship and time-dependent. Probiotics - the interaction of the cell-NDV change results of the cell culture supernatant of NDV hemagglutination monovalent NDV hemagglutination probiotics and the culture supernatant and the NDV simultaneously access the cells can be extremely significant reduction in price, suggesting that the kill the virus may have a direct role. In the experiment, we studied the probiotic viable culture supernatant, inactivated bacterial cells, and breaking the antiviral activity of the bacterial component, results showed that the various forms of the probiotic has a certain antiviral activity, but in the there are some differences in the effect may be due to the different mechanism of its antiviral activity. Probiotics cells and then infected with the virus, the result of the elevated NDV inhibition rate reflects the blocking effect of probiotics NDV adsorption cells; probiotics with the virus at the same time reducing access to the cells after the the virus cells infringement phenomenon, It can be seen that probiotics may have a direct damaging effects on virus structure; cells infected with the virus and then access probiotics NDV very low rate of inhibition phenomenon virus infection probiotics difficult to play a role. In summary, the The probiotic of NDV antiviral may be multi-faceted, with both the direct destructive effect on NDV blocking viral infection of cells, and can inhibit the proliferation of the virus in the cell.
