Alpinia officinarumHanee belongs to Zingiberaceae Alpinia Species,its root is used as medicine.It is one of the commonly used traditional Chinese medicine,which contained in 2005 Edition 1 of "Chinese Pharmacopoeia".It is warm in nature,pungent in flavour,can disperse cold and relieve pain, the main treatment is for the abdomen pain and vomit. Modern pharmacology is not only consistent with the traditional efficiency but also shows anti-inflammatory, antioxidant, antitumor, antibacterial and other new role. For more in-depth research on chemical components of Alpinia officinarumHanee, looking for the active substance, rational using its drug resources to promote the process of modernization of Chinese medicine, this work studied systematicly on the chemical components of the Alpinia officinarumHanee.This work use modern chromatographic techniques for systematicly separating ethanol extract from Alpinia officinarumHanee of Xuwen, and also analyzed the main active medicinal ingredients—Volatile Oil of Alpinia officinarumHanee.The volatile oil of A. officinarum Hance was extracted by hydrodistillation (HD), organic solvent extraction and ultrasonic assisted extraction. The optimum extraction process for volatile oil obtained by hydrodistillation method was investigated by the simple factor experiment.Afterwards,the chemical constituents of volatile oil were separated and identified by gas chromatography-mass spectrometry(GC-MS).The relative content of each component was determined by area normalization.The efficiency and chemical constituents of organic solvent extraction and ultrasonic assisted extraction were compared with that of traditional hydrodistillation. In addition, preliminary exploration about the effect of volatile oil on the preservation of aquatic products was done.The results showed that as follows:1.In the hydrodistillation extraction of volatile oil from Alpinia officinarumHanee, the effect of extraction time on the extraction rate of volatile oil was highly significant, crush degree of herb was significant and soak time was not significant. 2.The optimum extraction process for volatile oil obtained by hydrodistillation was as follows:the proportion of material to solvent is 1:10,crush degree of herb is 80 mesh,soak time is 2h and extraction time is 7h. In this extraction condition, extraction rate of volatile oil was(0.342±0.011)mL/g.3.Comprehensive comparison,ultrasonic assisted extraction method was more timesaving, detected more compositions, and this method can extract some volatile components which can not been extracted by hydrodistillation extraction.Thus, hydrodistillation extraction has some shortages, organic solvent extraction and ultrasonic assisted extraction can be used as effective supplementary methods.4.In this study, flavonoids in Alpinia officinarumHanee separated and purified by CTLC. After filtrating,select expanding agent as CH2Cl2:CH3OH=98:2, the flux of expanding agent was 3ml/min, collected by small centrifuge tube ,each 1.5 ml.The whole elution process was about 50 min. Compared with the traditional column chromatography separation,CTLC saved more time, used less eluent and more completely purified. It was determined that,in this study, galangin contained in Alpinia officinarumHanee collected from Xuwen was 6.89mg/g. Purified by CTLC,the extraction rate was 46.69%, purity was 89.2%. Flavonoids extracted in Alpinia officinarumHanee through the UV spectrum,infrared spectrometry and melting point analysis, confirming three substances which were Galangin, Kaempferide and Quercetin.5.Through sensory evaluation,the mensuration of TVB-N value and PH value,the result shows that, the volatile oil of Alpinia officinarum Hance can inhibit the changes of TVB-N value and PH value,maintain the sensory quality. Through the quantitative analysis,ordinary ice fish resulting in slightly smelly in 4 days, other indicators are all exceed the minimum limit value of fresh fish.The time of keeping fresh of tilapia stored in volatile oil of Alpinia officinarum Hance continued 8 days, indicators are all within the allowable range. Compared with the blank group, Preservative group can be extended at least 3 days or more.
