Abstract


Objective: To study the impact baicalin rabbit lens epithelial cell proliferation and expression of NF-κB , preliminary explore the baicalin to alleviate posterior capsular opacification role , to open up a new avenue for posterior capsular opacification medication . Methods : 1 rabbit lens epithelial cells in primary culture and subculture : the healthy eye diseases in adult rabbits killed venous air embolism , take the eye anterior capsule cut into a size of 1mm × 1mm debris interception seeded in flasks , Ⅰ collagenase digestion , the medium was changed once every 2 - 3 days , about two weeks after the cells covered most of the bottom of the bottle . 1:2 passaged with 0.25% trypsin digested , the choice of 2 - 3 generations of cells used in the experiment . Establishment of different concentrations , the concentration of drug in the different duration of action of the drug group and blank control group 3.MTT detected each time the rabbit lens epithelial cell inhibition rate measured by flow cytometry at 48h early apoptosis rate of various concentrations of semi-quantitative reverse transcriptase polymerase chain reaction (RT-PCR) to detect different concentrations of baicalin role rabbit lens epithelial cells , NF - κB p65 subunit of expression . Results : 1.MTT results showed that baicalin can inhibit the proliferation of rabbit lens epithelial cells , the inhibition rate with time , the concentration was positively correlated . Half of 48 hours the suppression concentration for 46.32μg/m1 2 . Flow cytometry testing confirmed baicalin can effectively induce apoptosis of rabbit lens epithelial cells , the rate of early apoptosis in a concentration dependent increments . 3.RT-PCR results showed that , with the blank control group , the baicalin role group NF-κB p65 mRNA expression was significantly reduced , and the difference was statistically significant ( P lt; 0.01 ) . Conclusion : baicalin can effectively inhibit the proliferation of rabbit lens epithelial cells induced apoptosis . Baicalin inhibition of rabbit lens epithelial cells may be related to NF-κB expression downregulation , NF-κB expression exception may be involved in the development of capsular opacification .
