Abstract


Acute phase reaction was considered as a non-specific innate immune mechanism, involved in defending inflammation and limiting microbial growth in response to outer stress. Three experiments were conducted to study the impact of some stress on the concentration of acute phase proteins (APP), and further evaluate the capacity of APP in assessing swinery health. It was expected to provide some experimental basis and theoretical direction on well knowing the swine real-time health status and the environment of pig farms.Experiment 1:Selecting and determinating APP index in blood for assessing pigs’ health. In order to select an appropriate method for assessing pigs’health, the fluorescence quantitative PCR (FQ-PCR) technology for APPs mRNA expression level and the enzyme-linked immune-sorbent assay (ELISA) for APP protein contents were compared. Using selected method assessed haptoglobin (HPT), major acute phase protein (MAP) apolipoprotein-Al (Apo-Al), C-reactive protein (CRP) and ai-acid glycoprotein (α1-AGP) concentration in blood, then some sensitive indexes or combinations were screened out. The results showed:ELISA was a more effective method for APP in assessing pigs’health; HPT, MAP and Apo-Al were more sensitive in indicating the body state than CRP andα1-AGP.Experiment 2:Effects of different treatments on productive performance, blood cell parameters and the APP contents for pigs. Experimental pigs were randomly assigned to 5 treatments, challenged intramuscularly with Hanks’balanced aslt solution (the control group), Pasteurella Multocide Vaccine (Live) (the PMVL group), Swine Japanese Encephalitis Vccine (Live) (the SJEVL group), the triple vaccine (Live) (the TVL group) and LPS endotox (the LPS group) respectively. The results were as follows:(1) In contrast to the PMVL group, ADFI in the LPS group was lower (P<0.05) in the first week. (2) The parameters of WBC, RBC and PLT (the mean platelet volume was an exception) in the PMVL group showed an earlier raised and later decreased trend. Nevertheless, the parameters of red blood cells and platelet in the SJEVL group, the TVL group and the LPS group decreased to some extent, and the WBC increased. (3) The concentrations of APP were related to the type and severity of the stimulations. The highest levels of the positive APPs (MAP and HPT) and the lowest levels of the negative APPs (Apo-Al) were observed in pigs with bacterial infection at 48 h and 72 h respectively. Whereas, the concentrations of MAP, HPT and Apo-Al reached the maximum or minimum at 24 h,72 h and 48 h respectively. (4) Regarding the APP response, a significant correlation(r=0.922, P<0.01) between the PMVL group and the TVL group were found. The effect of the SJEVL on APP was in accordance with reports, which concluded that the SJEVL group imitated viral infection successfully.Experiment 3:Relations of acute phase protein index for swinery health. The contents of HPT, MAP and Apo-Al in blood were determined by ELISA in this study. The results were as follows:(1) The concentrations of MAP (1.40+0.94μg/mL) and HPT (20.11±12.23μg/mL) in apparent healthy pigs were higher than that in apparent diseased pigs (1.23±0.70μg/mL,18.75±9.14μg/mL respectively), and 61.53±16.18μg/mL for Apo-Al in apparent healthy pigs was lower than that in apparent diseased pigs (69.24±13.49μg/mL). It was supposed that the concentration of APPs would tend to recover the initial values with time. (2) A significant correlation (r=0.999; P<0.01) was observed between MAP and HPT in blood, and Apo-Al and MAP had low correlation (r=0.252; P<0.05).In conclusion, the changes of APP could response the body health status, showing an association between the magnitude of changes in the APP levels and the type and severity of the stimulations. The concentration of positive APPs (MAP and HPT) in blood would increase significantly within 72 h after the body was infected, and the negative APPs (Apo-Al) decreased significantly. It was inferred that the concentration of APPs tended to recover the initial values with time.
