Abstract


Breast cancer is a common malignancy of women with breast cancer clinical onset age dropped lower and lower peak age of onset overall than Western women as early as 15 years, which greatly affect female health. In breast cancer and other malignancies, DLC1 inhibit cancer cell proliferation, migration and induction of apoptosis. MCF7 cells is a large, flat cells, polygonal, healthy state cell refraction is good, life cycle is short, vigorous, in this laboratory conditions to cultivate survive, is the study of breast cancer cells in good materials. Thus, this experiment MCF7 cells for the study. Flavonoids are present in most plants as a secondary metabolite, because they have anti-inflammatory, antioxidant, anti-tumor effect and attracted widespread attention. In order to study flavonoids tumor suppressor role of the molecular mechanisms of this experiment, flavonoids core structure: flavonoids (flavone) treatment of human breast cancer cells MCF7, design concentration gradient of flavonoids, with the MTT assay to determine the optimum flavonoid concentrations, further application of MTT detect flavonoids processing 48h, 72h and 96h after cell proliferation, with the scratch assay flavonoids before and after treatment of cell migration situation, and colony formation analysis and detection flavonoids on cancer clone formation using flow cytometry flavonoids on cell cycle distribution and apoptosis; using RT-PCR was used to detect flavonoids treated tumor suppressor gene DLC1 and cell cycle-related genes CyclinE and p21Waf1 expression. By MTT assay found 200μM flavonoids can effectively inhibit breast cancer cell proliferation, in order to further study inhibition of cell proliferation mechanism, flow cytometry flavonoid treatment on cell cycle distribution and found that flavonoids cell cycle gathered in the G0/G1 phase prevent cancer cells to S phase development, and to some extent induced apoptosis, thereby restrain cell proliferation. Semi-quantitative RT-PCR, flavonoids treated G1 phase cyclin CyclinE downregulation of cyclin-dependent kinase inhibitor p21 Waf1 upregulation, thereby blocking cell cycle progression, cell cycle arrest at G0/G1 phase, but not to S phase development, inhibit cell proliferation, restrain cancer, developmental role. Applications scratch test and tablet cloning experiments found that flavonoids can effectively inhibit cell migration and colony formation, in order to further study this effect the mechanism by semi-quantitative RT-PCR detected flavonoids treated tumor suppressor gene DLC1 expression, which regulates cell migration The ROCK genes downregulated, thus inhibiting cell migration activity. These results indicate that flavonoids by upregulating DLC1 gene expression, inhibition of cell proliferation, migration and colony formation, blocking cell cycle progression, to some extent induce apoptosis. Therefore, the use of appropriate flavonoids administered alone or combination therapy can prevent breast cancer lesions metastasis; in everyday life, food rich in flavonoids legumes can effectively reduce the risk of breast cancer.
