Abstract


Objective: animal studies for the treatment of chronic obstructive pulmonary disease, acute exacerbation of (AECOPD) cough particles by traditional Chinese medicine preparation, clinical efficacy and safety studies to explore the treatment of AECOPD mechanism cough particles and observe the clinical efficacy and safety. Its clinical development and application of clinical and experimental evidence. Methods: Animal experiments 1.1 take 60 SD male rats were randomly divided into six groups, each group of 10 were cough particles high group (K1), (K2), the low-dose group (K3), acute bronchitis syrup positive control group (A), model group (M) and blank control group (N). 1.2 method of administration: cough particles high, medium, low doses were administered 17.5g, 8.75g, 4.375g/kg, acute bronchitis syrup positive control group 20ml/kg blank control group administered saline 20ml/kg per once a day for consecutive 15 days. 1.3 SD of serum TNF-α, IL-8 was measured (using EILISA). 1.4 lung histopathological examination. Results: The serum levels of IL-8, TNF-expression of a 1.1 model group (M) and blank group (N), model group, serum IL-8, TNF-a expression rates, two sets of phase dose group and the control group of acute bronchitis syrup (A group than statistically significant (P lt; 0.01); 1.2 cough particles group K1 (17.5g/kg), K2 (8.75g/kg), K3 (4.375g/kg) ) compared of cough particles serum IL-8, TNF-a downregulated in the low-dose group with statistical significance (P lt; 0.05) high dose group was statistically significant (P lt; 0.01). HE staining observed by light microscopy results (see Figure 1-6) showed that the model successfully reproduced. Cough particles high in the low dose group to varying degrees to reduce the infiltration of inflammatory cells and reduce the production of mucus exudate, in which the high-dose group better. Conclusion: 1. Cough particles high in the low-dose group to reduce AECOPD serum IL-8 content, high dose group; 2. Cough particles in the low-dose group can reduce AECOPD rats serum content of TNF-α, in which the high-dose group more obvious; cough particles of three dose groups to varying degrees mitigate the AECOPD rat bronchial mucosa layer and submucosa infiltration of inflammatory cells, reduce the mucosal epithelial cell necrosis and shedding reduce airway secretions; 4. cough particles the AECOPD rats have a better therapeutic effect. Its anti-inflammatory mechanism may be related to reduced IL-8 and TNF-α expression. The clinical efficacy and safety observation 1) Selection of cases according to the 2008 GOLD guidelines to select AECOPD 60 patients, using the random number table is divided into: the control group and the treatment group; 2), treatment (see experimental methods). (1) control group: anti-infective, anti-asthma, cough and phlegm, anti-inflammatory, oxygen, or non-invasive ventilator and other symptomatic treatment; (2) in the treatment group: on the basis of the treatment in the control group serving cough particles. 3), laboratory detection of cases in both groups before treatment, after fasting blood samples were measured blood count, liver and renal function. Before and after treatment were measured lung function; 4), clinical observation of (see clinical observation). Results 1. Treatment and control groups before and after treatment, the clinical symptoms and signs score, with statistical significance (p lt; 0.05) (see Table 3); 2. Treatment and control groups before and after treatment, the blood count statistically significant (p lt; 0.05) (see Table 4); 3. before and after treatment of the control group and the treatment group liver, kidney function was not statistically significant (p gt; 0.05) (see Table 5-6); lung function between the treatment and control groups before and after treatment was statistically significant (p lt; 0.05) (see Table 7); Conclusion: The treatment group to improve AECOPDI clinical symptoms and signs score; 2 treatment groups can improve routine blood; 3 treatment groups can improve lung function; treatment group found no liver function, renal dysfunction, cough particles with security.
