Functions of Mitochondrial Biogenesis and Metabolism Related Genes in Mouse iPSCs Induction and Insight into Mechanism

Abstract


At present , the technology of inducing adult or other non-terminal differentiated somatic cells into pluripotent stem cells(iPSC) has matured. And some papers have reported that the iPSC can be generated from less than three or four factors (SKOM or SKO). However, being a new born technique, there are still many problems about the mechanism of reprogramming we still have not clearly understood, especially the central issue of the reprogramming mechanism is poorly understood. Mitochondria play a very important role in the process of iPSCs induction; besides, morphology and function of mitochondria have undergoing significant changes, and these changes could meet the need of iPSCs. Here, we generated iPSCs from mouse fibroblast for the analysis of mitochondria biogenesis related genes expression and its impact on the iPSCs reprogramming. On the one hand, during the process of iPSCs generation, mitochondria changes consistent with the iPSCs induction. On the other hand, changes of mitochondria biogenesis may enhance the iPSCs reprogramming. We screened a series of genes about glycolysis, oxidative phosphorylation and mitochondrial biogenesis, and try to find some genes can make some changes during the reprogramming. According to the experiments results, we found that the inhibition of mitochondrial biogenesis gene PGC-1 can enhance the efficiency of iPSCs generation. And, there are some proofs that the increase of the expression of PGC-1 could lower the iPSCs generation. Anyway, during the process of reprogramming of iPSCs, the expression of mitochondria related genes have become active.
