Objective: To study the effects of soluble vascular endothelial growth factor receptor 1 (soluble fims-like tyrosine kinase recepter1 of sFlt-1) differences in the levels of the protein, and serum sFlt-1 protein levels in normal and preeclampsia serum renal ball vascular endothelial cell monolayer permeability barrier, and to explore the relationship between preeclampsia serum sFlt-1 expression level changes in glomerular vascular endothelial cell injury. Method: 1.sFlt-1 protein expression level of determination: ELISA (enzyme-linked immunosorbent assay, ELISA) and quantitative detection of 12 cases of normal pregnancy, 12 patients with mild and 12 cases of severe preeclampsia serum sFlt-1 protein expression levels. 2. Glomerular vascular endothelial cells in culture and identification: Immunohistochemical detection of factor VIII of VEGF and CD31. 3. Glomerular vascular endothelial cell monolayer barrier function: Transwell chamber method described in 12 cases normal pregnancy, 12 patients with mild and severe preeclampsia serum intervention glomerular endothelial cells (human glomerular endothelial cell lines, GENC) growth of single fluorescently labeled bovine serum albumin detection glomerular endothelial cells layer permeability barrier function. The results: 1. SFlt-1 protein levels in the serum of preeclampsia than normal pregnancy group was significantly higher in severe preeclampsia serum sFlt-1 protein levels in mild preeclampsia group was significantly higher. Preeclampsia serum glomerular vascular endothelial cell function can be significantly impaired, and that damage to the barrier function of the glomerular vascular endothelial cells, elevated glomerular vascular endothelial cells by monolayer of bovine serum albumin concentration severe preeclampsia group, than mild preeclampsia bovine serum albumin concentration increased more obvious, that is the extent of the damage of glomerular endothelial cells heavier suits endogenous VEGF by single glomerular Bovine serum albumin concentration of vascular endothelial cells was significantly reduced. Dysfunction of glomerular endothelial cells in the degree of preeclampsia serum sFlt-1 protein levels positively correlated. Conclusion: preeclampsia serum glomerular vascular endothelial cell monolayer barrier permeability can be increased, may be associated with serum sFlt-1 antagonize VEGF role, the glomerular barrier function of endothelial cells is related to impaired. Objective: To study exogenous vascular endothelial growth factor (vascular endothelial growth factor, VEGF) in trophoblast cells sFlt-1 expression cause damage to the glomerular endothelial cells (GENC), from the cellular level to explore the sFlt-1 and VEGF in glomerular vascular endothelial cell injury in the role. Method: 1. Liposomes the sFlt-1/pcDNA3 plasmids transfected into trophoblasts by reverse transcriptase polymerase chain reaction (reverse transcription polymerase chain reaction, RT-PCR) and quantitative real-time polymerase chain reaction (real-time polymerase chain reaction, Real-Time PCR) detection sFlt-1 gene expression, ELISA assay supernatant sFlt-1 protein expression. 2. Supernatant of transfected group trophoblastic intervention glomerular endothelial cells using fluorescently labeled bovine serum albumin detection of glomerular vascular endothelial cell monolayer barrier function. Results: 1. Liposomes sFlt-1/pcDNA3 plasmid transfection to nourish cells 72h after transfection group of sFlt-1 protein levels and the expression of the gene level were significantly increased. Transfection group supernatant intervention glomerular vascular endothelial cell function was significantly impaired: the glomerular vascular endothelial cells by monolayer of bovine serum albumin concentration was significantly higher glomerular vascular endothelial cell monolayer permeability barrier function was significantly impaired, and the addition of exogenous VEGF bovine serum albumin concentrations were significantly reduced by the monolayer glomerular vascular endothelial cells. Conclusion: exogenous VEGF plays an important role in maintaining glomerular endothelial cell function. The trophoblasts of sFlt-1 overexpression can lead to glomerular vascular endothelial cell function was significantly impaired, which may induce preeclampsia kidney injury series of clinical manifestations of the important reasons.
