The Effect of IL-17 on VEGF Expression in Human Nsclc Cell in Vitro

Abstract


Objective To investigate the recombinant human interleukin- 1 (7recombinant human interleukin 17, rhIL-17) on cultured human non-small cell lung cancer cell lines A549 and H460 vascular endothelial growth factor (vascular endothelial growth factor, VEGF) expression levels. Methods not containing or containing different concentrations of rhIL-17 (0.1,1,10,100 ng / ml) in RPMI-1640 culture medium two cell lines A549 and H460 , then : (1) MTT assay rhIL various concentrations -17 for 48h and 72h on cell growth. ( 2 ) quantitative RT-PCR assay the role of rhIL-17 after A549 and H460 cells expressing VEGF mRNA levels. (3) ELISA was used to detect different concentrations of rhIL-17 A549 and H460 cells after stimulation of VEGF protein expression levels . The results showed that : ( 1 ) with different concentrations of rhIL-17 for 48h and 72h , A549 cells in vitro than the control group did not change significantly (P gt; 0.05); H460 cells after 48h rhIL-17 groups each concentration no significant changes in proliferation , and after 72h, 10 ng / ml and 100ng/ml rhIL-17 group was significantly inhibited proliferation (P lt; 0.05). ( 2 ) The concentration of rhIL-17 acts on the A549 and H460 , each group of VEGF mRNA and protein expression compared with the control group, the difference was not statistically significant. Conclusions ( 1 ) 0.1, 1 , and 100 ng / ml of rhIL-17 in vitro proliferation of A549 cells no direct role in promoting, but 10ng/ml and 100 ng / ml concentration of rhIL-17 for 72 can inhibit H460 cells in vitro . ( 2 ) In this study, various concentrations established by rhIL-17 in vitro against human non-small cell lung cancer cell VEGF mRNA and protein expression had no significant effect.
