Clinical Significance of MGMT and ERCC1 Expression in Gastric Carcinoma and Precancerous Lesion

Purpose by detecting DNA damage repair gene product-of MGMT of ERCC1 expression in gastric cancer and gastric dysplasia of MGMT to understand ERCC1 in gastric cancer development. Explore the possibility of MGMT and ERCC1 as a useful indicator for monitoring the biological behavior of gastric cancerous and judgment. Methods Immunohistochemistry SP method detected 16 cases of normal gastric mucosa, 18 patients with mild dysplasia, gastric mucosa, 24 cases of severe dysplasia in the gastric mucosa, 18 patients with early gastric cancer, 44 cases of advanced gastric cancer tissue expression of MGMT and ERCC1 . Results MGMT protein expression in the nucleus expression, part of the cytoplasm of a small amount of expression. MGMT positive expression rate in normal gastric mucosa, mild dysplasia of gastric mucosa in severe dysplasia and gastric mucosal early gastric cancer and advanced gastric carcinoma were 93.8% (15/16), 94.4% (17/18) 91.7% (22/24), 66.7% (12/18), 68.2% (30/44). MGMT protein expression in early gastric cancer rate was 66.7% (12/18) and the expression rate of advanced gastric cancer, 68.2% (30/44), significantly lower than the the mild dysplasia expression of the gastric mucosa rate 94.4% (17/18) compared statistically significant (P lt; 0.05). Gastric MGMT protein expression was, at the age of 60 (including the age group over 60 years old), the 60-year-old age group were 58.8% (20/34) and 78.6% (22/28), no significant differences between the two ( P gt; 0.05); in male and female patients were 67.4% (31/46) and 68.8% (11/16), between the two there was no significant difference (P gt; 0.05); diameter 5cm (5cm) more tumor group, less than 5 cm tumor group were 55.6% (10/18) and 72.7% (32/44), no significant differences (P gt; 0.05); were moderately differentiated carcinoma and poorly differentiated carcinoma 81.1% (30/37) and 48.0% (12/25), the former is significantly higher than the latter (P lt; 0.05); tumor infiltration depth of myometrial tumor reached serosa were 73.1% ( 19/26) and 63.9% (23/36), between the two was no significant difference (P gt; 0.05); tumors with lymph node metastasis and without lymph node metastasis were 59.5% (25/42) and 85.0% (17/20), the former significantly lower than the latter (P lt; 0.05). 2. ERCC1 distributed mainly expressed in the nucleus, also expressed in the portion in the cytoplasm, concentrated around the nuclear membrane. ERCC1 in normal gastric mucosa, mild dysplasia of gastric mucosa in severe dysplasia and gastric mucosal early gastric cancer and advanced gastric cancer tissue positive expression rate was 93.8% (15/16), 72.2% (13/18) 75.0% (18/24), 55.6% (10/18), 45.5% (20/44). ERCC1 expression in normal gastric mucosa group rate of 93.8% (15/16) was significantly higher than that of early gastric cancer was 55.6% (10/18) and advanced gastric cancer, 45.5% (20/44), the three statistically significant (P lt; 0.05). ERCC1 protein positive expression rate of stomach cancer at the age of 60 (age group, including more than 60 years old), the 60-year-old age group were 47.1% (16/34) and 50.0% (14/28), no significant difference between ( P gt; 0.05); between male and female patients were 47.8% (22/46) and 50.0% (8/16), no significant differences (P gt; 0.05); diameter 5cm (5cm) more tumor group, less than 5 cm tumor group were 55.6% (10/18) and 45.5% (20/44), no significant differences (P gt; 0.05); were moderately differentiated carcinoma and poorly differentiated carcinoma 59.5% (22/37) and 32.0% (8/25), the former is significantly higher than the latter (P lt; 0.05); tumor infiltration depth of myometrial tumor reached serosa were 69.2% ( 18/26) and 33.3% (12/36), the former is significantly higher than the latter (P lt; 0.05); lymph node metastasis and without lymph node metastasis tumors were 38.1% (16/42) and 70.0% (14/20), the former significantly lower than the latter (P lt; 0.05). 3.62 cases of gastric cancer tissues, MGMT and ERCC1 common positive in 26 cases, negative 16 cases together expressed unanimously by 42 cases the inconsistent 20 cases, 16 cases of MGMT positive ERCC1 negative, MGMT negative ERCC1-positive, 26 cases. Spearman rank correlation analysis, γs = 0.392, gastric MGMT expression and ERCC1 expression showed significant positive correlation (P lt; 0.05). The the conclusion 1.MGMT protein expression in early gastric cancer and advanced gastric cancer rate was significantly lower than the the mild dysplasia gastric mucosa (P lt; 0.05). ERCC1 expression in normal gastric mucosa group was significantly higher than early gastric cancer and advanced gastric cancer (P lt; 0.05). Tip MGMT and ERCC1 expression level of gastric carcinoma may represent the degree of DNA damage, may predict gastric malignant behavior of one of the indicators. 2.MGMT significant ERCC1 positive expression rate in patients with different tumor differentiation (well differentiated group than poorly differentiated group P lt; 0.05), positive lymph node metastasis rate lower than the group without lymph node metastasis positive rate (P lt; 0.05), and therefore the two indicators for the judgment of the degree of differentiation and lymph node metastasis of gastric cancer have a certain suggestive. With the progress of gastric mucosal lesions in precancerous lesions of MGMT and ERCC1 expression rates gradually declining trend both in gastric carcinoma were positively correlated (P lt; 0.05), suggesting that both the malignant process in the gastric mucosa play an important role as a useful indicator for monitoring of mucosal lesions progress and to determine the biological behavior, applied to the gastric mucosa follow-up testing of high-risk groups, in order to improve the detection rate of early gastric cancer. 4 simultaneous detection of the expression of MGMT protein ERCC1 protein, contribute to the early warning of gastric cancer.
