Abstract


China is a big country mariculture, the current density and intensive farming methods resulted in aquaculture water ammonia content seriously overweight, not only the culture water environment has been destroyed, returned to the coastal waters of serious environmental pollution and eutrophication phenomenon. Anammox bacteria under anaerobic conditions, the direct reaction of ammonia and nitrate nitrogen ability to reduce ammonia levels in the culture environment, maintaining farming ecosystem nitrogen cycle smoothly. To better understand the nitrogen polluting Jiaozhou Bay Red Island breeding areas anammox bacteria community structure and composition, the subject of the application hydrazine oxidoreductase gene (hydrazine-oxdizing enzyme, hzo) as a molecular marker of Jiaozhou Bay Red Island Aquaculture District anammox bacteria in the spatial distribution, diversity, phylogeny and abundance of in-depth research. Topic selection ponds inside and outside five stations were established anammox bacteria hzo gene clone library by restriction fragment length polymorphism (Restriction fragment length polymorphism, RFLP) analysis, a total of 625 clones was screened produce 104 operational taxonomic units (Operational taxonomic units, OTU), shows a higher diversity. hzo gene sequences in GenBank database, the similarity of the most similar sequences in 84% 1 00%. Diversity index analysis showed L3 stations anammox highest functional gene diversity, L2 station's ranking, while the lowest L4 stations. Each station Anammox community structure Jackknife Cluster and PCA clustering analysis showed that relatively clean environment, L1, L4, L5 three stations close Anammox community relations, while polluting L2, L3 stations population structure and the other three stations vary greatly, indicating Anammox in the spatial distribution of environmental conditions have a significant adaptability, also suggests using hzo gene as an indicator of environmental pollution and tracer. Integrated environmental factor clustering analysis revealed the Jiaozhou Bay Red Island culture environment Anammox community structure and sediment parameters are physical and chemical factors such as the result of a variety of conditions. The phylogenetic tree is divided into six clusters, culture zones anammox bacteria are mostly concentrated in the uncultured \Found in marine aquaculture environment, there are specific clusters with other clusters anammox bacteria low homology, and rarely found in other environments, it may be special circumstances to adapt to the unique culture zone cluster. L2 stations in the anammox bacteria hzo highest gene copy number (1.255 × 10 6 copies / g), L5 station lowest (1.257 × 10 5 copies / g), the result was consistent with the relative content, but also the L2 stations highest (1.1975 × 10 -5 ), L5 station lowest (1.3693 × 10 -6 ). Affect the number of copies distributed reasons yet to be studied by RNA transcript levels further revealed. This study reveals the breeding environment for the population structure of ammonia-oxidizing bacteria, ammonia oxidation process and the relationship between microbial species research is important, but also for aquaculture pollution control nitrogen in the environment to provide a theoretical basis.
