Diferential Proteomics of Leaf from Tobacco Varieties with Different Pressure Resistance

Abstract


Tobacco (Nicotiana Tobacco L.) is an important economic crops, occupies an important position in the national economy. Sustainable development of tobacco production is constrained by drought stress, at the same time by the tobacco black shank (Phytophthora nicotianae Breda de Haan) against. So far, the drought and black shank tobacco-induced gene expression, the molecular mechanism of the synthesis of specific proteins is unclear. In this study, proteomics research methods and techniques, the choice of four different resistance to the cultivation of tobacco varieties were used to extreme drought stress inoculation treatment or Phytophthora, exploratory study of tobacco leaves differential proteomics. The main progress as follows: 1, initially identified tobacco leaves total protein extraction method, the pH range of the IPG strip, protein sample volume experimental conditions. 2, drought-tolerant varieties of cultivated tobacco smoke 14 45 d after transplanting, and then after 12 d after drought stress from the proteome expression profiles, identified more than 300 proteins screened 52 different protein spots, 14 of which expression up-regulated the expression of 18 down, 15 is the induction of newly generated five protein spots disappear expression. After mass spectrometry and a variety of database search method successfully identified 42 (19) protein spots, of which: 31 (9) is related to the matter and energy metabolism proteins; 4 (4) antioxidant protein; (3) is a protein associated with the translation; 2 (2) is related to the signal transduction protein; 2 protein of unknown function ungrouped. Drought-sensitive varieties of cultivated tobacco smoke 100 45 days after transplanting, and then after 12 d after drought stress from the proteome expression profile of more than 330 protein spots identified, screened 43 different protein spots, of which 20 upregulation of the expression of 10 down, 12 is the induction of newly generated expression of a protein spots disappear. After mass spectrometry and a variety of database search method successfully identified 34 (14) differentially expressed proteins, of which: 28 (8) is related to the matter and energy metabolism proteins; 4 (4) antioxidant protein; (1) is related to the signal transduction protein; of an ungrouped protein. 4, a sense of black shank cultivated tobacco varieties 1025 small gold heel 3 weeks after vaccination Phytophthora spores treatment after 3 days from the proteome expression profile of about 260 protein spots identified, screened 38 different protein spots, of which 14 were up-regulated and 16 were down-regulated, 6 is the induction of newly generated two protein spots disappear. After mass spectrometry and a variety of database search method successfully identified 29 (15 species) differentially expressed proteins, of which 20 (9) is related to the matter and energy metabolism proteins; 1 (1) antioxidant protein; (3) is a protein associated with the translation; 1 (1) is related to the signal transduction protein; ungrouped protein 3 (2). 5, three weeks after the cultivation of tobacco 3 varieties innovations, heel resistance to Phytophthora Phytophthora spores inoculation treatment for 3 days, from the proteome expression profile of about 400 protein spots identified, screened 39 different protein spots, of which 14 were up-regulated and 20 were down-regulated, 3 induction of newly generated two protein spots disappear. After mass spectrometry and a variety of database search method successfully identified 28 (19) differentially expressed proteins, of which 15 (8) is related to the matter and energy metabolism proteins; 8 (7) antioxidant protein; (1) is related to the signal transduction protein; 2 (2) antiviral defense protein; 2 unknown proteins. Can be clearly identified by the analysis of the experimental results, the vast majority of the protein is involved in the matter and energy metabolism and resistance preliminary study of drought stress and inducing tobacco black shank resistance response mechanism. Lay the theoretical foundation for further screening breeding new resistant tobacco varieties resistant specific proteins as a marker character.
