Objective To investigate the expression of POKerythroid myeloid Ontogenic factor (Pokemon) and matrix metalloproteit-9 (MMP-9) in human glioma and meningioma tissue and to analyze their clinical significance.Methods According to WHO’s tumor classification of central nervous system in 2007, a total of 58 cases of glioma and 48 cases meningioma tissue were used to detect the expression of Pokemon and MMP-9 by immunohistochemical streptavidin-perosidase (SP) method,including 9 cases pilocytic astrocytoma and 5 cases astrocytoma (WHO gradeⅠ),8 cases astrocytoma and 3 cases oligodendrocytes astrocytoma and 1 cases oligodendroglioma and 2 cases ependymoma and 1 cases yellow multiforme astrocytoma (WHO gradeⅡ),1 cases astrocytoma and 4 cases anaplastic astrocytoma oligodendrocytes and 2 cases anaplastic astrocytoma oligodendrocytes and 1 cases oligodendrocytes astrocytoma and 3 cases anaplastic oligodendrocytes and 2 cases anaplastic ependymoma and 2 cases glioblastoma (WHO gradeⅢ),11 cases glioblastoma and 1 cases medulloblastoma and 2 cases glioblastoma multiforme (WHO gradeⅣ), and including 4 cases endothelial cell meningioma,4 cases transitional cell meningioma,4 cases fiber cell meningioma,2 cases hemangioma meningioma and 2 cases syncytial meningioma (WHO gradeⅠ), 16 cases atypical meningioma (WHO gradeⅡ),6 cases micro-malignant meningioma, 6 anaplastic malignant meningioma and 4 cases recurrent malignant meningioma (WHO gradeⅢ).Results1. Pokemon protein is expressed in different grade glioma and invasive meningioma samples.With the increasing of tumour malignant or invasive level,the positive expression rate of Pokemon also increased.The positive rate which in glioma was28.57%、66.67%、80.00% and 92.86% respectively, and the expression rate in meningioma was 25.00%、68.75% and 93.75%respectively. The positive expression rate of Pokemon was significant difference (P<0.05) among the WHO gradeⅠ, WHO greadⅡ, WHO gradeⅢglioma and meningioma tissues and WHO gradeⅣ glioma tissues.2. MMP-9 protein is expressed in different grade glioma and invasive meningioma samples.With the increasing of tumour malignant or invasive level, the positive expression rate of MMP-9 also increased.The positive rate which in glioma was 21.43%、53.33%、86.67% and 92.86% respectively,and the expression rate in meningioma was 14.29%、56.25% and 93.75%respectively. The positive expression rate of MMP-9 was significant difference (P<0.05) among the WHO gradeⅠ, WHO greadⅡ, WHO gradeⅢglioma and meningioma tissues and WHO gradeⅣglioma tissues.3. The expression rate of Pokemon correlated well with MMP-9 level (r=0.730, p=0.000<0.05)(r=0.881, p=0.000<0.05),with the increasing of expression of Pokemon the expression of MMP-9 also increased.Conclusion The expression of Pokemon and MMP-9 correlate with the clinical pathological grade, probably promotes the oncogenesis、progression and invasiveness of human gliomas and meningiomas, Detect its expression, can help determine the degree of malignancy of brain tumors and the development process for the future gene therapy of glioma and meningioma provide a basis to open up new areas of brain tumor treatment.
