Abstract


Objective: To detect prostasin in epithelial ovarian cancer patients before and after surgery serum levels and the corresponding pathological tissue prostasin expression and analyze their relevance to explore prostasin as a tumor marker CA-125 alone or combined test for ovarian cancer early diagnosis, monitoring treatment, prognostic value. Method: 1. Double antibody sandwich ELISA assay prostasin, CA-125 in 26 cases of epithelial ovarian cancer patients before and after surgery serum samples and 40 cases of benign ovarian tumors and 30 healthy female serum samples concentration. Investigate ovarian cancer patients with preoperative serum prostasin tissue protein concentration correlation between the expression intensity. 2 immunohistochemistry (immunohistochemical technique) detected in 20 cases of normal ovarian prostasin, 25 cases of benign ovarian epithelial tumors, 19 cases of borderline ovarian tumors and 56 cases of epithelial ovarian cancer in patients with resected tissue specimens, comparing four kinds of tissues prostasin expression, as well as prostasin in ovarian cancer tissues with different positive expression rate of the relationship between clinical and pathological features. Results: 1. Ovarian cancer group preoperative serum prostasin (14.07μg/ml) compared with healthy female group (5.66μg/ml), epithelial ovarian benign tumor group (7.85μg/ml) significantly increased, the difference was statistically significant ( P lt; 0.0.01), healthy women with benign tumor group compared no significant difference (P gt; 0.05). Ovarian cancer group were compared with preoperative serum levels of prostasin significantly lower (P = 0.000). (2) benign tumor group for women with healthy reference population, prostasin concentration 8.245μg/ml time, ROC-AUC was 0.867, (sensitivity specificity) -1 biggest difference, ie the best cutoff point. prostasin ovarian cancer detection sensitivity 84.6%, specificity was 81.4%; joint CA-125 detection sensitivity 96.73% and specificity of 100%. Ovarian cancer patients with preoperative serum concentrations of tissue prostasin prostasin protein expression was positively correlated (P lt; 0.05). 3. Prostasin in normal ovarian tissue was weakly positive or no expression in ovarian benign, borderline and malignant epithelial tumors, the positive expression rate of 24.00% (6/25), 36.84% (7/19), 71.47% (40/56), benign and borderline tumors showed no significant difference between the expression (P gt; 0.05); malignant and benign, borderline pairwise, expressing a significant difference (P lt; 0.001). Prostasin in ovarian cancer tissues with FIGO stage, histological grade, serum concentrations were positively correlated (P lt; 0.05). 4 in epithelial ovarian cancer group, preoperative serum and tissue concentrations prostasin expression of prostasin was a linear correlation. Conclusions: 1. Epithelial ovarian cancer patients preoperative serum prostasin concentration was significantly increased after surgery prostasin concentration decreased significantly. The later clinical stage, the higher the concentration of serum prostasin, suggesting that serum prostasin of epithelial ovarian cancer, clinical diagnosis, treatment and prognosis judgment has clinical significance; serum prostasin concentration does not depend on the CA-125 levels, suggesting that the two In the diagnosis of ovarian cancer can play a good complementary roles. 2. Prostasin in epithelial ovarian cancer tissues was significantly higher than the normal control group, benign epithelial ovarian tumors and borderline ovarian tumors, the positive expression rate of histological grade and clinical stage significantly associated with the pathological type, patient age , no significant relationship between menopause, suggesting that detection of epithelial ovarian cancer prostasin determine the degree of malignancy has clinical significance.
