Abstract


Objective: Expression and clinical significance of hyaluronic acid in non-Hodgkin's lymphoma, further research on CEM cells in vitro proliferation and invasion of exogenous hyaluronic acid, the prognosis of non-Hodgkin's lymphoma, the disease progress to provide reference; non-Hodgkin's lymphoma has opened a new anti-cancer targets provide a theoretical basis for clinical treatment. Method: 1. Peripheral venous blood samples collected 51 patients with NHL, separation of plasma and radioimmunoassay determination of plasma concentration of hyaluronic acid, gelatin zymography determination of the activity of plasma matrix metalloproteinase 2,9. Other plasma samples collected from 15 healthy people as controls. 2 containing different concentrations of hyaluronic acid role CEM 0ug/ml, 10ug/ml 20ug/ml exogenous hyaluronic acid 10? S 1640 CEM cell culture medium, determination of the following indicators: ⑴ MTT method measured cells 24, 48 h and 72h after changes in cell proliferation; trypan blue exclusion staining count different concentrations of hyaluronic acid role 24,48,72,96,120 h after the number of living cells CEM cells, and cell growth curve; ⑵ invasion Experimental analysis of different concentrations of hyaluronic acid role CEM cells 24 hours after cell invasion changes; ⑶ real-time quantitative PCR method to measure the different concentrations of hyaluronic acid role CEM cells 24 hours after cell MMP2, MMP9 mRNA expression changes; ⑷ ??gelatin zymography Determination of the role of different concentrations of hyaluronic acid CEM cells 24 hours after the cells of MMP2, MMP9 activity changes. Results: NHL plasma HA expression levels: initial treatment group, relapsed or refractory group were significantly higher than the normal control group (P lt; 0.05); naive group, relapsed or refractory group were significantly higher complete remission group (P lt; 0.05); complete remission group and normal control group showed no significant difference (P gt; 0.05); naive group with relapsed or refractory group showed no significant difference (P gt; 0.05). The initial treatment of NHL III and IV patients plasma HA expression levels were significantly higher than the I, Phase II plasma HA expression levels (P lt; 0.05). Plasma HA expression of MMP2 expression is positively related to the correlation coefficient r1 = 0.678, P lt; 0.01. HA expression of MMP9 expression in the plasma is also a positive correlation, the correlation coefficient r2 = 0.654, P lt; 0.01. Exogenous hyaluronic acid treatment CEM cell proliferation after 24h without significantly enhanced to handle more than 48h proliferation was markedly enhanced. 5. Exogenous hyaluronic acid for 24h CEM cells invasive ability was significantly enhanced. 6. Exogenous hyaluronic acid for 24h, CEM cells of MMP2, MMP9 mRNA upregulation of MMP2, MMP9 protein increased activity, and a dose-dependent manner. Conclusion: 1. Hyaluronic acid in the plasma of patients with NHL high expression and in NHL progression and clinical stage; NHL plasma hyaluronic acid expression of MMP2, MMP9 activity enzymatic linear positive correlation; prompted the high expression of hyaluronic acid in the plasma of patients with NHL, NHL, invasion and metastasis. Exogenous hyaluronic acid to enhance CEM cell lines in vitro invasion capacity, the mechanism may be raised CEM cell lines of MMP2 and hyaluronic acid of MMP9 mRNA expression and enhancements of MMP2 and MMP9 activity.
