Abstract


Objective: Ovarian cancer is a common malignancy of the female reproductive system. Ovarian malignancy in about 90% of epithelial ovarian cancer. Due to the relationship between the growth pattern and location of epithelial ovarian cancer, early diagnosis is difficult and prone to metastasis and widely planted, 5-year survival rate is very low, only 25% -30%. The pathogenesis of ovarian cancer is still not clear to some level of gene expression changes may be caused by multiple factors. KISS-1 gene is a recently discovered metastasis suppressor factor. Some studies suggest that the KISS-1 gene expression in many malignant tissues is reduced or missing, and may be closely related to tumor progression and metastasis. clear. Recent studies prove that OPN diagnosis of breast cancer, colon cancer, pancreatic cancer, lung cancer and prostate cancer, the disease monitoring and prognostic significance. However, the relationship of the KISS-1 gene and the expression of OPN and ovarian cancer, as well as whether the correlation, both to promote the occurrence and development of ovarian cancer reported domestic. This study aims to to immunohistochemical staining explore KISS-1 gene and OPN expression in ovarian cancer tissues and its clinical significance, as well as the correlation of the KISS-1 gene and the expression of OPN. Methods: Using immunohistochemistry (immunohistochemistry, IHC) SP method detected 43 cases of ovarian carcinoma, 24 patients with benign ovarian tumor KISS-1 gene and protein expression of OPN. 2 Statistical analysis: application SPSS13.0 statistical software for statistical analysis of data. P ≤ 0.05 was statistically significant difference, P ≤ 0.01 for the difference was significant statistically. Results: The relationship between KISS-1 gene expression and clinical parameters KISS-1 gene protein expression in 43 cases of ovarian cancer and 24 cases of benign ovarian tumor: KISS-1 gene protein by immunohistochemistry assay positive expression rate was 39.53% (17/43) in ovarian cancer tissues was significantly lower than in the benign ovarian tumor positive expression rate of 75.00% (18/24), both statistically significant difference (χ ~ 2 = 7.765, P = 0.005, P lt; 0.01). KISS-1 gene expression in ovarian cancer: expression of lymph node metastasis than those without lymph node metastasis group, the difference was statistically significant (χ ~~ 2 = 4.036, P = 0.045, P lt; 0.05); in different clinical staging group's expression, the expression of early cancer (I, II ovarian cancer) than late cancer (III and IV ovarian cancer) the difference was statistically significant (χ to 2 = 6.029, P = 0.014, P lt; 0.05) ; in different pathological type of ovarian cancer and different age of the group, KISS-1 gene expression no significant difference (P gt; 0.05). The relationship between the 2 OPN expression and clinical parameters detected by immunohistochemical method OPN protein expression in 43 cases of ovarian cancer and 24 cases of benign ovarian tumor: OPN protein in ovarian cancer tissue positive expression rate was 74.42 % (32/43) was significantly higher than that in benign ovarian tumor positive expression rate of 37.50% (11/24), both statistically significant difference (χ to 2 = 8.841, P = 0.003, P lt; 0.01). Expression of OPN in ovarian cancer: lymph node metastasis group was higher than that without lymph node metastasis, the difference was statistically significant (χ to 2 = 11.692, P = 0.001, P lt; 0.01); groups in different clinical stages expression, early cancer (I, II ovarian cancer) was lower than that of advanced cancer (Ⅲ, Ⅳ ovarian cancer) the difference was statistically significant (χ to 2 = 12.029, P = 0.001, P lt; 0.01); ovarian cancer different pathological types and different prevalence age group, the expression of OPN was no significant differences (P gt; 0.05). Immunohistochemical protein expression levels, KISS-1 gene and the expression of OPN the 3 KISS-1 gene and OPN expression in ovarian cancer was a negative correlation (r = -0.507, P = 0.001, P lt; 0.01). Conclusion: 1 KISS-1 gene was highly expressed in benign ovarian tissue was significantly higher than its expression in ovarian cancer tissue, the difference was statistically significant, suggesting that the KISS-1 gene may be involved in the inhibition of tumorigenesis, development and invasion and metastasis; OPN was highly expressed in ovarian cancer tissue was significantly higher than that in benign ovarian tissue expression, a statistically significant difference, suggesting that OPN may be involved in promoting tumor occurrence, development and invasion and metastasis. 2 KISS-1 gene in ovarian cancer metastasis, lymph node metastasis group was lower than that without lymph node metastasis, the difference was statistically significant; different clinical stage group, the expression of early cancer tissue (I, II ovarian cancer) higher than that of advanced cancer (Ⅲ, Ⅳ ovarian cancer), the difference was statistically significant. Prompted KISS-1 gene and ovarian cancer lymph node metastasis and clinical stage are closely related, the KISS-1 gene expression is reduced or missing to be able to promote the invasion and metastasis of ovarian cancer. KISS-1 gene expression in different pathological type of ovarian cancer and the different prevalence age group no significant difference. Tip OPN ovarian cancer lymph node metastasis and clinical stage is closely related to elevated expression of OPN can promote ovarian cancer invasion and metastasis. Sick in different pathological type of ovarian cancer and different age group, no significant difference in the expression of OPN. Prompted OPN tumor type and age has little sick. 3 KISS-1 gene and OPN expression in ovarian cancer tissues showed the opposite trend, suggesting that the two opposite role in ovarian cancer development and invasion, metastasis, and between the two may be related participate in the same mechanism.
