Occupies a very important position in the field of drug analysis , HPLC (High Performance Liquid Chromatography, HPLC) , and the column is the key to achieve effective separation . Therefore , the development of new types of chromatography media has important practical significance . The polymer monolith has the following advantages , such as the preparation method is simple , good acid and alkali resistance , the section can be changed by changing the proportion of the porogen, monomer and a crosslinking agent to the stationary phase structure and pore size distribution , in the separation of biological macromolecules It has certain advantages. Therefore, the main contents of this paper is to develop a new type of monolithic column , and as the structure and performance of the chromatographic medium -depth research . Firstly, the overall classification of the column , preparation and application to do a preliminary introduction and a brief introduction of the advantages of hybrid materials . Secondly, the use of the atom radical transfer technology , ferrous chloride and carbon tetrachloride catalytic system to the development of a new type of organic - inorganic hybrid monolithic column . The monolithic column is based on glycidyl methacrylate as the monomer, ethylene glycol dimethacrylate as the crosslinking agent , dodecyl alcohol and methanol as a hole-making agent , ferrous chloride and carbon tetrachloride as a catalytic system to made of a hybrid monolithic column . And examines some of the performance of the whole column , permeability and mechanical stability of the polymer materials were investigated , and the results showed that the overall column has good mechanical endurance . Also inspect the whole column as the stationary phase high performance liquid chromatography . Application of the monolithic column lysozyme standard separation and separation conditions were preliminary optimization . Finally, the use of new hybrid monolithic column prepared , of ovalbumin standard separation conditions were explored , preliminary optimization of separation conditions . And using the determined separation conditions ovalbumin actual sample eggs were separated , the ideal separation results .
