Abstract


Isolated and cultured rat intestinal microvascular endothelial cells in vitro , and the immunocytochemistry were identified . Results: under an inverted microscope observation, cells were typical paving stone -like , non- overlapping growth phenomenon ; VIII factor related antigen Once positive endothelial cells . To investigate the role of lactic acid inhibit LPS - activated NF-κB pathway endothelial cells of cultured rat intestinal mucosa as model cells after different concentrations of lactic acid pretreatment with LPS attack using the ELISA method to detect the inflammatory cytokines TNF- α and IL-6 levels , and , respectively, from the level of genes and proteins to detect the expression of NF-κB p65 subunit . The results show that the lactic acid can be very significantly reduced TNF-α and IL-6 levels ; peak of TNF - α the expression in the first nine hours , the peak of IL - 6 The expression of the first 12h , the high concentrations of acid pretreatment group of inflammatory factor expression compared with LPS positive control group difference was very significant , and low concentration of lactic acid pretreatment group was a significant difference compared with LPS positive control group ; lactic acid of high , medium and low concentrations of NF-κB mRNA expression 1.51 times the control of the blank group , and 2.62 times , 3.00 times , and the NF-κB mRNA expression amount of LPS group was 7.36 times that of the blank control group ; each group protein extracts exists in both a molecular weight of about 65kDa , with the rabbit anti NF-κB P65 polyclonal antibody immunoreactivity in protein bands , and the semi-quantitative results show the NF-κB P65 expression in the cells of lactic acid pretreatment group compared with the blank control group difference was not statistically significant ( P gt ; 0.05) ; LPS control group, NF - κB P65 expression and other groups compared to the differences were highly significant ( P lt; 0.01 ) .
